Mesenchymal stem cells in human bone marrow contribute to haematopoiosis. Since mesenchymal stem cells have characteristic adherence to plastic, they can be isolated from the bone marrow. And MSCs have been characterized phenotypically as non-haematopoietic cells, as they do not express CD34 or CD45. Human MSCs can be identified through flow cytometry by the monoclonalantibodies. They show extensive proliferative capacity in vitro without the loss of phenotype and retain the ability to differentiate into several mesenchymal lineages. MSCs constitutively secrete a large number of cytokines and promote the expansion and differentiation of haematopoietic stem cells(HSCs) in vitro. It is generally accepted that bone marrow mesenchymal stem cells (MSCs) can trigger low responsiveness of allogeneic T lymphocytes against alloantigens in vitro, which can alleviate GVHD after transplantation, but whether at the same time weaken GVL effect? It is worthing considering. Objective: To address the question whether allogeneic T lymphocytesinteracted with mesenchymal stem cells from bone marrow still have killingactivity, investigate the feassible strategy for preventing GVHD whilepreserving GVL effect.Method:Activised donor T lymphocytes were co-cultured with bone marrowMSCs from patients. The killing activity with different effect/taget ratioof co-cultured T lymphocytes to primary leukemia cells were evaluated withLDH assay.Result: The killing activity to primary leukemia cells of allogeneic Tlymphocytes co-cultured with bone marrow MSCs reduced obviously comparedwith control(T lymphocytes un-cocultured with bone marrow MSCs) . Thekilling activity of 25:1 effect/taget ratio decreased obviously whencompared with that of 50:1 effect/taget ratio.Conclusion: Allogeneic T lymphocytes co-cultured with bone marrow MSCstillhad killing tumor cells ability, but the killing activity decreased.
