Scar is the product of tissue repairing. Scar can influence the appearance of skin. The contracture of scar also can cause the disorder of joint moving. So scar will bring the un - convenience to burning patients after healing. Most of the scar after burning is hyperplasia. It grows to the spike in 6 months after healing. Many studies prove that the degree of hyperplasia and contrature of scar is connected with man-y agents and the most important is the un - proportional increase of fi-broblast and the super deposition of collagen. Many eytokine can influence the hyperplasia and cntracture of scar and the most important is transforming growth factor β ( TGF - β) in them. It can stimulate hyperplasia and contracture of scar in many ways. It' s content is in direct ratio to the degree of hyperplasia. And γinterferon is the cytokine that has negative influence to hyperplasia of scar and has the opposite action to TGF -β. In this experiment , we use cell culture method to observe the influence of γIFN to TGF - β in hyperplasia scar. So we could prove the inhibition action of γIFN to scar. We also can find the proper concentration of this action.MaterialsWe select 10 patients including 8 men and 2 women and the mean age is 32 years old. The scar is from 0. 5 to 2 years after injuring. The scar is higher and harder than normal skin. It is no red swelling and ulcer. And they never have been treated with physical or chemical method. When the scar is resected, they are preserved in low temperature refrigerator ( - 80℃).MethodsAfter being sterilized, the scars are digested to remove the other components and only reserve the fibroblast to undertake the primary culture. When the primary culture is finished, we use the same number of fibroblast to undertake culture again. There are different concentration of γIFN (0U/ml, 10U/ml, 100U/ml, 1000U/ml) in the culture fluid. When this culture is finished, we fetch the supennatant fluid to detect the concentration of TGF -β with EUSA method and the values are analyzed statistically. Then we can know weather γIFN has the inhibiting action to TGF - β and the proper concentration of γIFN with this action.ResultsAfter the experiment, we can know that there is no statistically difference between 10U/ml group and 0U/ml group. And there is obvious statistical difference between 100U/ml,1000U/ml groups and0U/ml group. But there is no statistical difference between these two groups.DiscussionsScar is the dys - repairing product of skin. The degree of hyper-plasia and contracture of scar is connected to many factors. And the most important is connected with the dis - equilibrium of collagen metabolism. In hyperplasia scar, the synthesis of collagen is increased and the amount of collagenase is decreased. At the same time, the ratio of III type collagen is increased. The degree of contracture of scar is connected with the number of myofibroblast. The hyperplasia of scar is also connected with the change of extra cellular matrix. TGF - β play very important role in the hyperplasia of scar through many ways. First, it can increase the number of fibroblast and stimulate fibroblast to secrete collagen. It also can decrease the speed of degeneration of collagen through inhibiting the action of collagenase. So it can stimulate hyperplasia of scar obviously. Second, TGF - β, can induce the expression of asoomth muscle actin in fibroblast. So the number of myofibroblast would be increased and contracture of scar is produced. At last, it can stimulate fibroblast to produce extra cellular matrix and delay its renewal speed. The degeneration speed of collagen is also relieved. In hyperplasia scar, the amount of TGF -β is increased obviously and its amount is in direct ratio to the degree of hyperplasia of scar. Therefore, we can say that TGF -β play a important role in hyperplasia and contracture of scar.
