Diabetic mellitus is chronic disease. The angiopathy and neurop-athy are principal complications. Among these complications the neu-ropathy is very general. There is no evident symptom in neuropathy during early diabetic period. It appeares hypoesthesia, spontaneous pain, muscular hypotonus induced by the change of motor nerve, my-atrophy or palsy and dropping blepharon induced the lesion of cranial nerve, diplopia. There are some alterations of pathology including the decrease of myelinated nerve fiber after light microscopic examination and the dilataltion of microtubule in axis - cylinder, degeneration of myelin sheath, the dissolve of structure plate layer, regeneration, cel-lular proliferation of Schwann.Main blood vascular smooth muscle is innervated by adrenergic nerve. With the thorough investigation, the vasodilator is founded, in-cluding nitroxidergic nerve and peptidergic nerve. We have proved the vasodilator in hindlimb of rats, and the neurotransmitter might be cal-citonin gene - related peptide. The diabetic patient often goes with hy-pertension, and the prevalence rate is 30 -40%. Animal experiment shows that the role of vasodilatation on skin induced by nerve is weak-ened. The result indicates that vasodilator might have pathologic chan-ges. In this study, we have observed the function of vasodilator has changed in diabetic rats induced by STZ throught electric stimulation in spinal cord of pithed rats.MethodsThe STZ rat model is set up by i. p. STZ solutions(60mg/kg). After pithing, methoxameine and hexamethonium are input through vein by constant current pump. Then the rat receives electric stimula-tion in spine cord, and the stimulation is repeated during five mi-nutes. The vasodilation effect is expressed by the fraction of the im-proved mean blood pressure. The result is expressed by x ?s. Signifi-cant assay uses T - test, and the value of P <0. 05 is considered sig-nificant.ResultsThe blood glucose is improved and the weight is lighted with time passing by in STZ diabetic rats. The blood glucose has no change and the weight increases in experimental control group. The blood glucose and weight have no difference in experimental control and blank con-trol group. The success rate of diabetic model by injecting STZ is 82.7%.The mean blood pressure and heart rate descents after pithing in experimental and control group. When rats receives electric stimula-tion in spine cord, their blood pressure decrease . The vasodilation fraction had no difference in experimental and experimental control group during 4W and 8W period. But, it shows significant difference during 12W period. The effect of electric stimulation has no significant difference in experimental control and blank control group.DiscussionThe STZ rat model is used commonly experimental diabetic ani-mal model. In this study, we successfully induce experimental diabet-ic model by using STZ(60mg/kg, i. p. ). The induced diabetes corre-spond with type 1 diabetes.In this study, we block transmitter of sympathetic nerve in pithed rats, to contract the vascular, and then electric stimulation can induce blood pressure decrease. This result implies that there are vasodilator in response to rats with electric stimulation in spinal cord.It's reported that myelinated nerve fiber of 8W diabetic rats have showed atrophied and thinned. 11W diabetic rats have both myelin sheath lesion and axis - cylinder lesion. Those alterations can induce nerve reaction weaken or extinction. The result of diabetic rats in dif-ferent periods (4W,8W,12W) shows that there is no significant difference between diabetic rat (4W and 8W) and control group, but there is significant difference between 12 W diabetic rats and control rats.It indicates that perivascular vasodilator may change and have re-duced function with the elongtion of diabetes .It has reported that experimental diabetic rat has endothelium -dependent and endothelium - independent vasodilation response re-duction.
