For this reason,it has important significance for clinical surrounding - operation period to know the damage - ma-chine of adhesive molecular in obstructic jaundic and how to decrease its damage.Cell adhesive molecular is a categy of glycoproteins of membran surface. It has a important role in the many physiologic and pathologic process of inflammatory reaction and immunity response. Intercellular adhesive molecular -1 (ICAM - 1) is more important and the mark of leucocyte activation.. Recent investigations have maken clear; the damaging action of ICAM -1 is that it can facilitate a large amount of leucocyte to collect in the liver sinus, then release flammatory medium and cause organ damaged. Therefore, and - adhesiveness therapy can release organ damage, but there are not operative techniques.Recent years, scholars has paid close attention on growth hormone in the treatment of obstructive jaundic. It has been clear by our pro-phase investigations that GH may release endotoxin and TNF level by improving intestinal mucosal machinery barrier of obstructive jaundic. But this machine is not reported and should be investigated.This study will discuss the machine and influence for rhGH to the expression of ICAM - 1 and procee experimential exploration for clince.Materials and MethodsExperiment animals: adult male wistar mice 45 body weight 300 -350 grams.Methods: 1. Dividing the 45 mice into three groups in random and average A: sham operation group ( SO ) , 15; B: obstructive jaundice group(OJ) 15;C: OJ + rhGH group, 15.2. The mice are fixed on the operation table, and oncised with knife to open the abdominal cavity. Then we will dispose the hepatodu-odenal igament to find the common bile duct. The mice in B group and C group suffer from bile duct ligation, while the control griup not. After doing all these, we close the abdominal cavity with thread. 3. Twelve hours later the mice are allowed to eat and drink freely and move free-ly.4. Two weeks later OJ +rhGH group mice is injected with rhGH subcutaneously in the posterior leg once a day. The dose is 0. 75iu/ 100g,and the other two groups are injected with saline just the same dose.5. Three weeks later the animals are killed and liver and blood are gathered.6. The plasm is taken out of blood to detect the bilirubin in the three groups.7. Liver are composed to tissue section with HE stain and ICAM -1 stain,which are examined under light microscope.ResultThe manifestation of jaundice in group OJ and OJ + rhGH oc-curred after 1 week or so , with significantly higher level of TBIL and DBIL than that in group SO . .In group OJ the liver is swelling and fi-brotic, it feels very hard and nodularlike on the surface. The common bile duct is enormously enlarged just above the ligation part. The walls feel very thin tension. This difference showed the successful build -up of OJ model.Under light microscope the focal necrosis of hepatocyte can be seen in the tissue section with HE stain in group OJ, well in group SO the formation of liver is normal. The result of 1C AM - 1 stain : group OJis significantly higher than that in group SO, two groups numerb a-dopt q check - out;q > q0.01 > P < 0. 01;group OJ + rhGH is signifi-cantly lower than that in group OJ,q > q P <0. 05.DiscussionRecently, studies have detected that the chang of adhesive molec-ular is correlated to postoperative infection and multi - organs failure of obstructic jaundic.
