PrefaceIn our country hepatocellular carcinoma ( HCC) is one of the most malignant types of tumors. The morality of this disease ranks second of all the malignant tumors, and the disease has severely affect the people 's health. In the comment times , the exact mechanisnm is not very clear. Nowdays, the study of transcription factor E2f and Bel - 2 apop-tosis off gene is becoming new hot point. E2f can control many expression of the gene which control the proliferation of cell, including onco-gene etc. E2f can up regulation or down regulation of cell growth, these two functions are dependent on different tissue. So there are some discussions between the function of E2f. Bel - 2 is the production of B -cell lymphoma gene. Bel - 2 can restrain the apoptosis of cell. It can make cell exist longer and induce the carcinoma. Today in country there are no report about the function and relation of E2f, Bel - 2 in hepatocellular carcinoma.Material and Method74 HCC and 15 cirrhosis cases studied were seen at the China Medicine College Secondary clinic hospital, during the period of 1992 - 2001. All liver species were fixed with buffered formalin and embedded in paraffin. All the carcinoma species have the no carcinoma tis-sue. Immunohistochemical staining was performed on 5 -m - thick formalin fixed paraffin embedded tissue sections based on the standard , indirect strepeavidinbiotin horseradish peroxidase method using diaminodenzidine as a substrate, statistical analysis use Chi - square test and correlation analysis.ResultThe expression of E2f-3 is located in plasm and nucleus, which was brown particles. The expression is local and widespread. The expression of E2f-3 inHCC was 78. 4% ,and in para - carcinomar tissue is 28.4% ,the comparison between two tissue is significant (p <0. 05) The expression of E2f - 3 in HCC is78. 4% , and in cirrhosis tiaaue is 26. 3% ,the comparison is significant. The expression of E2f-3 is related with histologic grade,tumor size( p <0.05) ,but isnt related with gender, HbsAg, numder of tumor, histologic type, capsule, TIPV and AFP ( p > 0. 05 ). The expression of Bel - 2 is located in plasm,which is chtyso - brown particles. The expression is local or widespread. The expression of Bel -2 in HCC is 79.2% and in para -carcinoomar tissue is 24. 3%. The comparison is significant. The expression of Bel -2 in HCC is 79. 2% in cirrhosis tissue is 66. 2% ,the comparison isnt significant ( p >0.05) ,the expression of Bel -2 is related with histologic grade, tumor size, tumor number ( p < 0. 05 ) , but it isnt related with gender,HbsAg,capsule,histologic type,TIPV and AFP( p >0. 05). Correlation analysis about the 74 HCC's expression score is r =0.5874 ,t <0.05, which means the correlation between E2f-3 and Bel-2.ConclusionE2f transfer factor was found in adenovirus at first. It play th transcription function to E2 start, nowdays have already found 5 members of E2f family from E2f -1 to E2f -5. There is hight homology among these members, their function almost is the same. E2f include many homology rank,which is located in the gene's upper stream that play the important function from G0 phase to S phase. The target gene of E2f indued DNA sythesis and cell cycle gene,oncogene. but E2f also can induce cell apoptosis. Some report that E2f can induce cancer such as the high expression of E2f can induce fibroblasst of rat embryo into cancer. At the same time, there are high expression of E2f in car-cinomar of stomach, cancer of colon, myeloma, lung neuroendocrine turner etc human malignant tumor. But some reseacher reported the rat which is with the deletion of E2f function. These rat's thymus and lymph node issignificant enlarged. From these report believed that E2f can inhibit tumor. In our experiment E2f is high expression in HCC, but is lower in para - carcinoma tissue, cirrosis tissue and the expression is related with tumor grade, tumor size.
