Bronchial asthma is a chronic inflammation of airway in which all kinds of inflammatory cells enjoined, such as eosinophil, mastocyte, lymphocyte. By the metamorphosis reaction, allergen can activate a serial of immune cells, which can release the inflammatory medium to form the inflammatory reaction and hyperresponsiveness of airway. In addition, nervous system plays an important role in bronchial asthma by way of creating nervous cytokines to be involved in the attack of brochial asthma.NF - KB is a kind of important protein of nucleus transcription factors, it can regulate lots of the expression of cytokines and increase the number of genes' product of these cytokines. Under the normal condition, NF - KB factor is inactive in the cytoplasm with its inhibitors I KB, while it can be activated by outer stimulus ( such as inflammatory medium^ virus and antigen) ,then it can translocate from cytoplasm to nucleus and bind with promoter or enhancer of the corresponding inflammatory medium genes to increase the expression ofthese medium' s gene. Nowdays, although hormone was applied to cure the bronchial asthma clinically, it demands further discussion that whether it' s mechanism is related to the inhibition of NF - KB. This experiment want to explore that NF - KB play an important role in attack of bronchial asthma and the mechanism of hormone' s curing bronchial asthma by way of studying NF - KB activity in lymphocytes of micro - airway and neurons of C7 - T5 spinal ganglia in guinea pigs which were in asthma and treated by hormone respectively.Methods1. Foundation of bronchial asthma model24 guinea pigs (200 - 220g, all female. ) were divided into 4 groups random, 1ml N. S was injected into abdominal cavity of guinea pigs of N. S group(4) ,then inhaled the mist of N. S after lOdaysjlml 10% OVA was injected into abdominal cavity of guinea pigs of simply allergized group(4) ,then inhaled the mist of N. S after 10 days (two groups above were control groups) ; 1ml 10% OVA was injected into abdominal cavity of guinea pigs of bronchial asthma group ( 8 ) , then inhaled the mist of 1% OVA after 10 days. Stopped inhaling after bronchial asthma reaction emerged and relieve spontaneously. And hormone group ( 8 ) had the same operation as bronchial asthma group, after that, dexamethsone(2mg/kg/d)was injected into the abdominal cavity of guinea pigs. Inhaling mist of guinea pigs in each group carried out once per day and killed the guinea pigs after 10 days.2. Drawing the materials, Embedding, SlicingGuinea pigs were piped and fixed routinely, then drawed the C7- T5 spinal ganglia and lungs, throwed them into PFA to post - fix for 24 hours, took them out and put them into 30% saccharu buffer to sink. Embedded them by OCT. Sliced by cryostat,the thickness of tissue slides was 12um. The slides had immunohistochemistry stain.3. Immunohistochemistry stainWe had immunohistochemistry stain through applying NF - KB rabbit anti - mouse antibody ( anti - P65,1:200, diluted by 0. 5% Tri-tonX -100) and SABC kit and DAB.4 . Statistics processWe measured the gray scale of the result by using Luzex - F real time picture analysis system. The data was processed by SPSS.Results1. In N. S group, the epithelia of micro - airway was intact with few of lymphocytes in bronchial wall, The NF - KB positive products which were scattered in the cytoplasm of lymphocytes were more than the ones in nucleus . The NF - KB positive products in the neurons of C7 - T5 spinal ganglia were almost scattered in cytoplasm, however, the positive products rarely scattered in nucleus. There was significant difference between the stain of nucleus and cytoplasm of lymphocytes and neurons by statistics ( P < 0.05 ) .2. In simply allergized group, the epithelia of micro - airway was intact and there was few of lymphocytes in the bronchial wall. And the NF - KB positive products which were scattered in the cytoplasm of lymphocytes were more than the ones in nucleus. The NF - KB positive products in the neurons of C7 -...
