Objectives: To clarify the current situation of wild measles virus of Xinjiang and increase information for China measles virus gene bank; simultaneously to establish mechanisms for reference for measles laboratory to diagnose measles; then to provide background information for measles control and eradication in Xinjiang. Methods : We collected throat swabs and urine specimens from the susceptibilities to measles; then we isolated measles virus by culturing the specimens into B95a cell line which is sensitive to measles virus; finally, we identified the genotype of the virus with RT-PCR technique. Results: Totally 4 wild measles viruses were isolated after we cultured the 42 throat swabs into B95a cell, and the positive rate was 9.52% (4/42 ) ; No virus was isolated from urine specimens; 3 of the 4 viruses were collected in Urumqi city, the other one came from Miquan city of Changji prefecture, and no measles virus was isolated from the specimens of other prefectures. Conclusion: The four isolates belong to H1 genotype and H1 genotype virus was the leading cause of measles in Xinjiang recently. Throat swabs must be sent to laboratory within 48 hours after being collected. Urine specimens is not suitable to the condition of Xinjiang ,we suggest that throat swab is the only specimen to be collected in the future. Methods of this project can be used in the measles virus laboratory surveillance in the near future. It provides background information and reference to measles control and eradication in Xinjiang.
