| The occurrence and development of tumor are linked with disregulation of cell cycle.CDKs activated by cyclin D1 and cyclin E enable phosphorylation of key substrates such as the pRb and release of transcription factors,which initiates DNA replication and consequently enters the S phase. As one of the CKIs,p21cip1 can inhibit cell proliferation by inhibiting the activiaty of CDKs-cyclinD1/cyclinE complex. Overexpression of cyclin D1 and cyclin E play a role in the occurrence of tumor.The objective of this study are to detect the mRNA content of cyclin D1, cyclin E and p21cip1 in CRCC of pTNM I stage,and to investigate their biological significance by comparing to clinicopathologic feature of these tumors. METHODSRT-PCR was used to detect mRNA content of cyclin D1, cyclin E and p21cip1 in 23 CRCCs of pTNM I stage and 7 nomal renal tissue distant to tumor. Immunohistochemistry was used to investigate the expression of p53 and ki67 in pathological tissue sections of CRCCs.The relationshipbetween mRNA content of cyclin D1, cyclin E, p21cip1 in CRCCs and the age, gender, tumor size, necrosis, Fuhrman nuclear grade, the expression of p53 and ki67 were analyzed statistically. RESULTSIn CRCC,the mRNA content of cyclin D1, cyclin E and p21cip1 is 0.484±0.416, 0.472±0.340, 0.356±0.215, respectively,which are all higher than that of the control group(p-value is 0.000 , 0.000 , 0.046,respectively).The lower cyclin D1 content was related to tumor size <3cm(p=0.008),but the lower cyclin E was related to tumor necrosis(p=0.022) and higher nuclear grade(p=0.004),the lower p21cip1 to higher nuclear grade too. A positively correlation is present between p21cip1 and cyclin E(p=0.002).None of these mRNA content is related to age, gender, the expression of p53 and ki67. CONCLUSIONThe mRNA content of cyclin D1, cyclin E and p21cip1 are all higher in CRCC of pTNM I stage than that of the control group,which is possibly related to the occurrence and development of the tumors. |
