Effects of Monoflurophate on the Mandibules in Ovariectomized RatsObjective: The purpose of this study was to investigate the relationship between systemic osteoporosis and mandibule as well as the effects of monofluorphosphate and a D3 on the mandibules of ovariectomized rats for clinical practice.Methods: 39 female Sprague-Dawley rats about 90 days of age were randomly divided into the following treatment groups and treated for 12 weeks as follows:Sham. There were 9 sham-operated rats.OVX. There were 10 rats in this groups.Sham and OVX served as aging and ovx controls and received distilled water used for dissolution of MFP.MFP. Ten ovx rats were given monofluorphosphate for 6 days/week at a dose of 0. 45mgF- + 13. 56 mg Ca2+/kg/d-1 body weight, by gastric feeding at the begining of seven weeks after ovariectomy.MFP+αD3. Ten ovariectomized animals were received MFP as described above for group MFP. These rats were also treated withαD3 at a dose of 0. lug/kg/d-1 body weight.At 8 and 3 days before necropsy, all rats were given intraperitoned injections of demeclocycline and salcein usedas f luorochrome bone markers. The rats were sacrif ised by ether . The blood samples were collected for determination of biochemistry parameters. The right femur and the right mandible were collected and fixed in 10% phosphate-buff ered formalin for histomorphometry. The left mandibule and femur were preserved under -20≥ in refrigerator for three-point bending test.Results: CD Compared with sham group, ovariectomy was followed by an increase in body weight. No difference in body weight was observed between treated group and OVX group. (2) The volume of womb in OVX groups is obviously smaller than that of sham group. (3) The estradiol was obviously higher in the sham group than in the OVX group. The alkaline phosphatase was significantly greater in OVX group than in the sham group, however the alkaline phosphatase in MFP+@D3 group is similer to that of sham group. There is no significant difference in bone gla protein between any of the group. (4) The fluorochrame-labeling marker is obvious in OVX compared with sham group, while the treated group is similer to that of the sham group. (5) The bone area and bone thickness in the OVX group were significantly lower than those in sham group. Conversely, the trabecular separation of the OVX group was significantly higher than that of sham groups. The mass of bone increased dramaticly in treated group.
