Bacterial meningitis is still associated with a high mortality and neurological sequelae(5-10% and 20% respectively) in children although much more advanced antibiotics have been used. Recent studies have demonstrated that antibiotics lead to deterioration of brain edema and intracranial hypertension in short-term because of their dissolving effects on bacilli. Clinical data also demonstrated 85% of deaths with bacterial meningitis occurred within 16 hours after they were in hospital. In this experiment, rabbit was inoculated intracisternally with 2 * 107 colony-forming units (CPU) of antigen-Kt positive E. Coli in order to duplicate E. Coli bacterial meningitis model. QingKaiLing is a kind of combination drug derived from NiuHuangAnGong pilular. It has been used clinically broadly because of its multifunction. E. Coli bacterialmeningitis rabbits were treated with antibiotics that QKL was treated as adjunctive drug. We observed its effects on bacterial meningitis in order to demonstrate its neuroprotective effects and mechanisms. Section OneDuplicate E. Coli bacterial meningitis model in rabbits Objective: To duplicate model of bacterial meningitis of Escherichia coli (E.coli) of rabbits. Methods: Rabbits were inoculated intracisternally with 2><107 CPU of antigen-Ki positive E. Coli. We observed symptom and determined the number of leukocyte > , protein and lactate content of cerebrospinal fluid (CSF) of rabbits respectively. While the content of water, sodium, potassium, calcium of brain were determined at 26h after inoculated. The pathomorphism of cortex of left brain and left hippocampus portion of rabbits were examined. Results: 1 Symptom and sign: The occurrence of hyponoia. inappetence and hypertonia in meningitis group was significantly higher than control group at 16h after inoculation (p<0.01, p<0.05, p<0.05) . The occurance of fever in meningitis group was significantly higher than control group at 16h after inoculation(p<0.01).2 The number of leukocyte and the protein content of CSF: The number of leukocyte and the concentration of protein in CSF of meningitis group were all significantly higher than control group at 16h after inoculationIX(p<0.01) . 3. The lactate content of CSF: The lactate content of CSF of meningitis group was significantly higher than control group at 16h after inoculation (p<0.01) .4. The content of water and electrolyte of brain: The content of water and sodium and calcium of brain in meningitis group were all significantly higher than control group at 26h after inoculation (p<0.01) .While the content of potassium of brain was significantly lower than control group at 26h after inoculation (p<0.01) .5 Pathomorphism '.Morphologic: In meningitis group, brain of rabbits were swollen and we found congestive vessel in tensive meninges. Gyrus became fleeter and exudation of ventricle was excessive. Microscope: In meningitis group we found sparse neurocyte and diffuse focal edema and bigger clearance of neurocyte and congestive vessels in cortex of brain. Soakage inflammation cells were also found in soft meningeal and arachnoid antrum. We found disorganized neurocyte arrangement in hippocampus. Vacuole denaturalization and crimple nuclear and blurry cytoplasm in hippocampus were also found. Conclusion: Antigen-Ki positive E.
