0bjective: To exp1ore the laws of cell adhesionmolecules (s ICAM-1, sP-selectin) changing during inflammatoryresponse to cardiopulmonary bypass. Intervention Therapy withLigustrazine,To observate the changes of cell adhesion molecules andto evaluate the mechanism and clinical significance of ligustrazine,thus provide a new approach of Anti-adhesiveness therapy. Method:32 cases having final diagnosed as congenital heart disease weredivided into ligustrazine group (20 cases) and controlgroup (12 cases)in random. Ligustraz ine (5mg/kg) were added to extracorporealcirculat ion machine in Ligustrazine group. In each group arterialblood sampl es were taken at the following times after induction ofanesthes ia, 15 minutes after the start of CPB, and 2 hours, 6 hours,12 hours, 24 hours after CPB. sICAM-1, sP-selection were measured bydouble antibodies sandwich Enzyme-linked immunosorbent assay (ELISA),Test kit come from GeneMay Inc. San Diego. Results: Plasma levelsof both measured adhesion molecules in both groups at baseline werein norma1 raflges. After CPB, levels of adhes ion malecules weresignficantly incrased in both groups. (s ICAM-1 from 127. 02 ±15. 33ng/ml to 345. 77 ± 26. 62ng/ml soluble P-selection from 28. 78 ±1. 99ng/ml to 106. 50 ± 6. 63ng/ml). The plasma levels of sICAM-1,4sP--se1ectin 1n control group were higher than in l igustrazine group(P<0. 01). The p1asma 1eve1s of s1CAM--1, sP--selectln in CPB 1astedlonger than 60 minutes were significant1y higher than in CPB 1asted1ess than 60 minutes. Conc1usions: The p1asma leve1s of sICAM--1,sP--se1ecti n were si gni fi cant 1y increased dur ing inf1ammatoryresponse to cardiopu1monary bypass. Ligustrazifle can weaken theexpression ol' so1ub1e 1CAM--1, So1ub1e P--se1ect i n and re1 ieve theinf1ammatory response to CPB, thus Ligustrazi ne can decrease theoccurrence of Comp1 icatlon of open heart surgery.
