In order to explore the effects and mechanism of Chinese cobra venom serum of rabbit (CVSR) on proliferation and apoptosis in leukemia and lymphoma cells in vitro, serologic pharmacological test was used. The effects were observed by means of trypan blue dye exclusion test, flow cytometry analysis, agrose gel electrophoresis and immunohistochernistry. The results showed that: (1) HL60 cells were inhibited significantly by the dosage of 2.5mg/ml, 5.0 mg/ml, 7.5mg/ml of CVSR. At the same dosage, there is no influence of CVSR on proliferation of human embryo lung diploid cells. Furthermore, the Jurkat and Raji cells showed the same inhibition when the dosage was added to 15 mg/ml. (2) By flow cytometry analysis, we found that the apoptosis rate of HL60 cells was significantly increased by the CVSR. A characteristic DNA "ladder" was detected by using agarose gel electrophoresis. (3) Bcl-2 and C-myc gene protein expression of HL60 cells was down-regulated. The results suggested that: (1) CVSR inhibited the growth of leukemia and lymphoma cells but had no cytotoxin on human normal cells in vitro. (2) HL60 cells was more sensitive than Jurkat and Raji cells to the CVSR. (3) The gene protein expression of bcl-2 and C-myc on HL60 cells down-regulated by CVSR was probably related to apoptosis.
