Compared The Quality Of The Dermatophagoides Extracts From China By Immunochemical Techniques

[Objective] Using the immunochemical techniques to analyze the quality of dermatophagoides allergen extracts from different laboratory in our country and to find a series of techniques that fit in with the WHO standard and suit the quality control of the allergen extracts in our country. One of the allergen extracts is more approaching the WHO standard.[Methods] The compositions of the samples from different units were separated by SDS-PAGE according to the molecular weight. And then detected the antigenicity of each band by Western blotting. The total biologic potency of all the samples was analyzed and compared by ELISA.[ Results ] The samples from The second affiliated hospital of Guangzhou Medical College and The Shanghai Medical University were detected to get 3 bands respectively by SDS-PAGE. And their molecular weights were 14KD, 18KD, 25KD and 14KD, 20KD, 58KD individually. There was no band for the samples which from Peking Union Hospital and Wuchang Railway Hospital. The bands detected by SDS-PAGE were identified using Western blotting. Negative finding was got from the control by Western blotting. The standard curve of the standard sample from ALK was acquired by ELISA. The results of others were comparedwith the curve: None of the actual content of all samples was in coincidence with their labels. The antigenic activity ratio of the samples examined using the serum of the rabbit anti-mite was similar to the results using the patient serum. The activity of the sample from the second affiliated hospital of Guangzhou Medical College was highest.[Conclusion] The content and the compositions of major allergen and the total biologic potency of the extract of the house dust mite manufactured in our country were not coincidence with their labels. So the techniques of the allergen quality control in our country need to be improved. The methods in this study are an exact, simple and repeatable technology than that were used previously, and it is urgent to standardize the process of allergen in China.
