OBJECTIVE To study the effects of Bozhi on anti-virus and protecting myocardium cells infected with Coxsackie 63 virus(CVB3) in order to provide theoretical and experimental basis for therapy of viral myocarditis(VMC) in clinic. METHODS The model of VMC was constructed by inoculating CVB3 in newborn rats' myocardium cells with the concentration of 100TCID50. The experiment was divided into 4 groups. Group A:normal cells group;Group B:after adsorbing the cells with CVB3 for 2 hours,the cells were washed out and added to Bozhi solution;Group C:after adsorbing the cells with CVB3 for 2 hours,the cells were washed out and added to carbamidine hydrochloride(CH);Group D:virus-infected .group. It was determined by MTT method that the virulence and the inhibition of pathological changes of Bozhi and CH in virus-infected SD rats' myocardium cells. The morphology,pulsation and ultra-structure changes of myocardium cells were observed by light microscope and electronic microscope. It was determined by colorimetric method that the activity of succinate dehydrogenase (SDH),cellular lactate dehydrogenises (LDH),glutamate oxalacetate transaminase (GOT). It was determined by biochemistry that the activity of super oxide dismutase (SOD) and content of maldondialdehyde (MDA). The CVB3-RNA in virus-infected group and medicine administrated group was performed by PCR to study the anti-virus effects of Bozhi. RESULTS The results showed that (1) the Bozhi could efficaciously inhibit the pathological changes of morphology,pulsation and ultra-structure for themyocardium cells infected by CVB3;(2) improve the livability of myocardium cells infected by CVB3;(3) significantly reduce the content of LDH,GOT and MDA released by myocardium cells and increase the activity of SOD;(4) and as compared with virus-infected group,the value of OD0 in the Bozhi administrated group was obviously reduced. CONCLUSION The experiment results indicate that (1) the Bozhi has good protecting effects for the CVB3- infected SD rats' myocardium cells and low concentration of the Bozhi could improve the livability of myocardium cells;(2) Adjusting the activity of SOD and MDA,the Bozhi effectively eliminates lots of free radical produced by infection of virus and inhibits the propagation of virus in myocardial cells to anti-virus;(3) The model of myocardial cells cultured in vitro has the merit of having the same conditions,good stability and convenient operation to be used to the other studies for VMC. The Bozhi has the function of anti-virus effect and protecting heart muscle cells from CVB3-induced myocarditis in SD rats.
