Experimental study in repairing articular cartilage defects with transplanting separated autolous periosteumPostgraduate:Jiang zhe Advisor:Han dong riObjectve To explore the factors affect the repairing articular cartilage with joint transplanting the autologous separated periosteum and red bone marrow ,and study the process of dynamic morphological changes.Method Usuing 42 of the Chinese rabbits .the 3mm 6mm of articular genus by surgery. And four groups were established for the experiment according to the different transplant tissues: Group A was transplanted with separated autologous periosteum alone, Group B with red bone marrow and separated autolous periosteum. Group C with red bone marrow and macro-peiec of periosteum, Group D with no control .The animals were sacrificed by stages on the 7th days and 2nd ,4th,8th,12th weeks after after operation, and the morphological changes of transplanted au autoogous red bone marrow and sepatated autologous periostum were observed by lighte and electronic microscopy and technology of image analysis.Result The articular cartilage defects were completely repaired by joint transplanting of autologous periosteum and red bone marrow after 8 weeks of operation, every technological index reached to normal, cartilage around the original defect and whole cells were surrounded by collagenous fibril, there was statisticallysignificant difference between experiment group and control group and control group in the single cell area with variance analysis.The experiment results that (1) the transplantation of entire separated autologous periosteum, the complex tissue of red bone marrow and periosteum, red bone marrow and macto-piece of periosteum could Repair the articular cartilage defects;(2) the peci-order of articular chondroblastic ability in autologous transplantation tissue is the complex tissue of red bone marrow and macro-piece periosteum, complex tissue of red bone marrow and entire perio- steum.(3)the process of regenerating the articular cartilage by repairing tissue is divieded into multiply stage of cell diffe- rentiation,formation stage of immature cartilage, reconstruction stage of cartilage.(4)the cells took parting repairing the articular cartilage defects have the undifferentiated mesenchymal cells in red bone marrow and periosteum, bone marrow stromal cells and osteogenic cells.
