| Objective: As we all know,hypertension will have influence on various system in humen body.Many studies to hypertensive rats have shown that the spermatogenic epithium of testis will change abnormally even affect the male fertility with increasing of blood pressure.The present study is aimed at evaluating the mechanism of infertility during hypertension by investigating the expression of iNOS ,eNOS and ET in testis of Dahl hypertension rat.Technology of light microsocope, immunohistochemistry ,cell-culure and radio-immunity and so on were involved in this study.Background: NOS is the key enzyme of NO synthesis.There are two kinds of NOS which are original and induced types.The original type include nNOS and eNOS.The induced type is iNOS. NO is produced under the catalyzing of NOS.Generally the main pathological change of hypertension is the spasm and harden of arteries.Because of disfunction of endothelial cells ,NO is dificite, dilating of vessels is straint.NO have ambifunction,lower level can prompt cell growth;higher level can inhibit cell growth.Many experts found that inreproductive system ,lower level NO is involved in spermatogenesis and capacitation.But more NO will damage testis tissue, reduce blood flow, affect secretion of angrogen and spermatogenesis. ET is the most effective long-term vascular contracting factor which we have ever known.lt is contributed to regulate testicular blood amount,contraction of seminiferous tubule and transportion of spermatozoon. Recently we found the NO and ET in serum of Dahl rats is higher than nomal. So discussing the change of NOS and ET in testes of hypertensive rats have great significence in order to explaining the mechenism of spermatogenesis damage resulting from hypertension.Materials and methods: Animals were Dahl salt sensitive rats which were imported from Japan.They were allowed to drink and eat freely .Inbreeding was implemented strictly.The pure zygote offspring is senstive to salt,which is a good modal for the study of hypertension.Selecting male rats,High salt feed when 5 weeks old and the normal feed groups were chosen as control.The test groups selected 9,10,11,12,13 week-old rats and the old-matched rats as control groups (n=6).Testes were taken after being infused with 4% polyformaldehyde,embeded in paraffin and sectioned as usual for light microscope and immunohistochemistry study.Selecting 18-20 day-old normal and Dahl rats ,Inaseptic environment ,testes were taken and digested by enzymes.Then sertoli cells were gotten and inplanted in culture-medium,after four generations ,these cells were used for immunochemistry stain.Results: The blood pressure of test groups is higher and higher with the prolonged course of disease.Seminiferouse tubules were damaged significantly.Immunochemistry results as follows:The positive productions were brown granules and located at the plasma of the cells.In test groups ,the expression of iNOS in sertoli cells and leydig cells are stronger than control groups(p<0.01).The expression of iNOS in test groups is decresing with the higher of blood pressure. eNOS:The positive cells include vascular enditheliocytes,some sertoli cell and some leydig cells myoid cell in nomal testes.In test groups,besides above cells,some spermatogenic cells are also positive, the immunopositive stain is more extensive than normal testes(p<0.01).In addition ,the positive rate in cells took on increasing tendency as the disease progressed in test testes.(3)ET: Positive reaction was mainly in endothelium cells of vessels and some sertoli cell leydig cell of nomal groups.In hypertensive rats sertoli cell leydig cell and some endotheliocytes of vessels are positive.And the cells displayed more strongly positive expression which is increasing with the development of hypertension(P<0.01).In cultured sertoli cell of normal and Dahl rats ,immunopositive products of iNOS eNOS were all located in these cells,and the expression in sertoli cell of nomal and hypertensive rats have no significent difference.During radio-immunoassay,... |
