Qidanyishenchongji(QD) was developed by professor Hong chunzhan according to an effective traditional Chinese medicine formula treating glomerulonephritis,which is composed of Radix Astragali,Radix Salivae Miltorrhiza,Radix Notoginseng,Ramulus Cinnamomi,Rhizoma Alismatis,etc.. This experiment used cationized bovine serum albumin(C-BSA) to induce membranous glomerulonephritis of rats and observed the pharmacological action of QD,and discussed the possible therapeutic mechanism.50 male Wistar rats (weight 150-200g) were randomly devided into 5 groups:high dosage of QD treated group (QD1 group),low dosage of QD treated group(QD2 group) ,shenyanshu treated group(C group),normal control group(N group) and model control group(M group).There are 10 rats in every group. Except N group,other rats received subcutaneous injection of C-BSA 0.5mg(dissolved in 0.5ml physiological saline and equivalent Freund's adjuvant incomplete) at back and axillas as previous immunization. A week later,these 40 rats received C-BSA l.Smg (dissolved in 1ml physiological saline) intravenously in the tail veins 3 times a week as formal immunization,9 times all together. From the third week after formal immunization,QD1 group were administered QD at the dose of 8g/kg by oral daily,QD2 group were administered QD at the dose of 4g/kg by oral daily,and C group were administered shenyanshu at the dose of Ig/kg by oraldaily,N and M groups received 20ml/kg physiological saline per day. At the second week after formal immunization,protainuria qualitative determination were examined weekly,And from the third week ,urine was obtained by housing individual rats in metabolic cages for 24hours to measure urinary protein At the 6th week all rats were sacrificed,blood and kidney specimen were collected to measure serum albumin,Scr,BUN ,TC,TG,and to observe histomorphologic changes under light microscope ,electron microscope and fluorescence microscope.20 mouses (10 male and 10 female) were administrated 0.6g/ml of QD,4 times daily. After 7 days observation ,maximum tolerance dose was calculated.Results:!.24h urinary protein rat the end of the third week,N group showed significant difference compared to other 4 groups (P<0.01),it means glomerulonephritis model were made successfully.From the 5th week,QDgroups showed significant difference compared to M group(P<0.01).From the 6th week,QD groups and C group all showed significant difference,it means QD can reduce urinary protein. 2. Serum albumin(ALB):QD groups showed significant difference compared to M group (P<0.01,P<0.05),it means QD can increase ALB level.3.Serum creatinine (Scr)and serum urinary nitrogen(BUN):Both Scr and BUN of QD groups showed significant difference compared to that of M group(P<0.01).C group showed significant difference (P<0.05),it means QD can protect renal function.4. Serum cholesterol (TC) and serum tryglyceride(TG):TC of QD groups showed significant difference compared to M group(P<0.01).C group showed significant difference (P<0.05).TG of QD groups and C group allshowed significant difference(P<0.05),it means QD can modulate lipometabolism.5. histomorphologic change:in M group,glomeruli were swelling ,GBM were thicken irregularly,foot process were merged together,IgG deposited along glomerular capillary wall in a "granulous" pattern ,fluorescent intensity were between +++ and++++.In QD groups,glomerular swelling was slighter than in M group,GBM were thicken occasionally,foot process merged partially,there are subepithelial electron-dense deposits,fluorescent intensity was weaker than M group or presented negative. In C group pathologic damage degree was between M group and QD groups. 6.Mouses maximum tolerance dose was 133.33 times compared with human's dosage.lt means QD is almost no toxicity.To sum up,QD is a effective preparation of traditional Chinese medicine,it can reduce proteinuria significantly,improve the renal function and hyperlipemia,alleviate pathologic damage.
