A Preliminary Experimental Study On The Antimutagenic,Antineoplastic And Immunoregulatory Activity Of Six Kinds Of Chinese Medicinal Herbs

PurposerTest the antimutagenic , antineoplastic and immunoregulatory activity and mechanism of six kinds of Chinese medicinal herbs(CMH) include hawthorn , mushroom , apricot kerneK jujube , isatis rooU and hairyvein agrimony, produce scientific evidence for tumor prevention.MethodsrThe antimutagenicity and mutagenicity assay of six kinds of CMH were investigated using the antimutagenicity and mutagenicity synchronous test.Vitamin C(Vit C) was used as antimutagenic positive contrast medium, mitom-ycin C(MMC) was used as mutagenic positive contrast medi-um,normal saline(NS) was used as neither antimutagenic nor mutagenic negative contrast medium,the test was divided into two parts:with and without rat liver microsomal enzymes (S9).The antineoplastic activity of the six kinds of CMH to four kinds of tumor cells line(MT-2,L929,SKV20 and K562) were investigated using in vitro antitumor test.In this test, phosphate buffered saline(PBS) was used as negative contrast medium,cisplatin was used as positive contrast mediu-m,tumor cells were cultured with CMH or contrast medium at 37癈 for 48 hours in 5% CO2 and 95% air,then the cellswere labeled with 3H-TdR for the last 24 hours and were harvested with the aid of a semiautomated cell harvester,the amount of radioactivity incorporated into DNA in the cells was measured with a liquid scintillation counter.Effects of the six kinds of CMH on proliferation of spleen cells of mice were investigated using 3H incorporation method.PBS was used as negative contrast medium,concanavalin A(con A) was used as T cell specific metogen, Lipopolysaccharide (LPS)was used as B cell specific metogen, spleen cells of mice were cultured with CMH or contrast medium at 37癈 for 48 hours in 5% CO2 and 95% air, then the cells were labeled with 3H-TdR for the last 24 hours and were harvested.The component of hawthorn and hairyvein agrimony were investigated by chromatography.Results: 1.With and without S9 tests both showed that the six kinds of CMH had no mutagenicity,hawthorn, apricot kernek jujube and mushroom were antimutagenic against MMC.2.Four different concentration groups of hawthorn , mushroom and hairyvein agrimony extracts all had the suppressive activity for proliferation of four kinds of cells(P<0.01),the inhibition activity had an increasing tendency with the increasing of concentration.Only the highest concentration group of apricot kernel(4mg/ml) had the suppressive activity for proliferation of L929 cell(P<0.01), only the highest concentration group of isatisroot extract(4mg/ml) had the suppressive activity for proliferation of L929 and SKV20 cells(P<0.05)jujube extract didn't show evident effects of inhibiting the proliferation of four kinds of cells.3.The six kinds of medicinal herbs showed the effects of promoting the proliferative activity of spleen cells of mrce,but the appropriate concentration of the six kinds of medicinal herbs are different.4.By means of chromatography showed that the main component of hawthorn were polysaccharide and small parts of protein,hairyvein agrimony were polysaccharide and protein.Conclusion :Hawthorn , apricot kernel -, jujube and mushroom have antimutagenic activity; hawthonu mushroom and hairyvein agrimony can apparently inhibit the synthesis of DNA of four kinds of cells(MT-2,L929,SKV20,K562);all the six kinds of medicinal herbs have the effects of promoting the proliferation of spleen cells of mice;the main component of hawthorn were polysaccharide and small parts of protein,hairyvein agrimony were polysaccharide and protein.
