This study was supported by the Fujian Provincial Natural Science Foundation for the theory of noninvasive photodynamic localization of Nasopharyngeal Carcinoma (NPC) and the Fujian Provincial Education Department key Science and Technology project for the medical instrument of laser dynamically diagnosing nasopharyngeal carcinoma, respectively. The major works in this thesis was focused on how to develop the drug-fluorescence imaging method for diagnosing and localization of early NPC.Firstly, it shows that it is very important to improve the early diagnosis rate of NPC in order to gain better treatment results. Furthermore, the feasibility for the diagnosis and localization of early NPC by drug-fluorescence imaging method was confirmed.Secondly, Experiments have been performed to study the spectral characteristics of the traditional photosensitizer HpD as well as the new photosensitizer PsD-007, HMME and ZnPcS2P2 in the physiological saline with 10 percent serum and in the pure physiological saline, respectively. Experimental results show that HMME and ZnPcS2P2 are the optimum photosensitizer for Photodynamic Diagnosis (PDD) and Photodynamic Therapy (PDT) for NPC, respectively.Next, the theoretical analysis of the drug-fluorescence optical imaging system for diagnosis and localization of early NPC is presented in detail from the point of view of laser and optical techniques, which mainly including the commercial nasal endoscopeIVand image intensifier. The fundamental optical parameters of transfer lens group were then ascertained and optical path calculation was made to adjust the aberration at the paraxial point. As a result, the definition and contrast of the fluorescence image by using drug-fluorescence optical imaging system can permit the detection and localization of early NPC. The optical design of laser transfer system mainly involves the coupling between light source and optical fiber as well as the direct coupling of two coaxial optical beams from different optical fiber without contact.Finally, optical phantom with different concentration photosensitizer was used to simulate NPC lesion in our performance test of the drug-fluorescence optical imaging system, and the preliminary results show that the system can meet with the need for the clinical diagnosis and localization of NPC. However, clinical trials should be carried our in the near future after further improvement on the performance of the drug-fluorescence optical imaging system.
