Objective: The prescription of compound Danshen Root has been applied for lots of cardiac or brain vascular diseases widely since 70s in 20th century. Though it possesses clear curative effect; many preparations; high sales quantities and thorough basic research, the ratio of Danshen Root and Sanchi in all products is different. Filtrating the best ratio becomes the body of our research. During the process of matching regulation research, we will supply the further constitutes matching with technological platform and methodology basis. So we can supply scientific warrants for developing modern compound herb prescriptions. Method: We apply the classical model-the left coronary artery ligated in canines resulting in the model of acute myocardial infraction and combine with hemodynamic model organically. With a view to whole constitutes and vivo animals and development observing, establishing the increase-decrease experimental design method of baseline geometric proportion, we carry through the comparative pharmacodynamics research on 7 ratios between Danshen root and Sanchi: 10/0,10/1,10/3,10/6,10/10,1/10,0/10 by many indexes. Effect evaluation indexes involve the electrocardiogram of outside velum of heart ( -ST, N-ST), the infraction scope of myocardial tissue, myocardial biochemistry markers(cTnI, CK-MB), coronary blood flow, myocardial consume the oxygen, hemodynamics, Nitric Oxide, Endo Thelin , free radicle, etc. Make it become the typical model of small sample, high dimension numbers, and many domino effects. At the same time we adopt the fuzzy technique with the artificial intelligence to cany through integrated judge about pharmacodynamics experiment data. Result:1. Influence on myocardial infraction: these groups which Danshen root is main, namely 10/6,10/3,10/1,10/0, are obviously able to ameliorate canine myocardialinfraction and alleviate the infraction degree from outside velum of electrocardiogram of heart and then reduce infraction section that TTC show when dying. Among them 10/6,10/3 is more outstanding.2. Influence on myocordial biochemistry markers: these groups which Danshen root is main make a restraint to the rise of cTnl obviously. We can say that the direct effect of Danshen root on cardiovascular is more prominent than that of Sanchi. The point is consistent with the result of electrocardiogram and myocardial infraction when dyinj.3. Influence on coronary blood flow and cardiac oxygen consumption: Some groups (10/0,10/3,19/6) which Danshen root is main are stronger than other groups about CBF. Western medic ire group takes effect after 30 minutes given the medicine, then function die down very quick, but the Chinese herbal grou;.:s (10/0,10/3,10/6) go into effect steadily and longer while rising constantly. So long-dated effect is obvious.4. Influence on hemodynamics indexes: Given the medicine, every Chinese herbal comparison groups take effect certainly. In a word, 10/6,10/3 groups can obviously ameliorate systolic and diastolic function of left ventricular; increase the cardiac output and the cardiac index; decrease the total periphery resistance but the left ventricular work index have no obvious augment while heart rate and mean arterial pressure have no obvious fluctuating.5. Influence on Nitric Oxide, Endo Thelin, free radicles: on all accounts. 0/10 group (pure Sanchi) not only promote the release of NO and protect the activity of SOD, but also lower the content of ET and reduce the production of MDA. There are stronger biological activities on these four indexes.6. Adopting fuzzy mathematics and clustering analysis, we sum up 10/6 and 10/3 are the superior groups by quantitative and qualitative analysis. The results are basic consistent with what we obtain by groovy methods.Conclusion:1. Danshen Root acts on vas as targets mostly; it expands coronary arteries more evidently than Sanchi.2. Sanchi acts on cardiac muscle as targets mostly; it prevents cardiac muscle from lacking in oxygen more evidently than Danshen Root.3.
