Objective To investigate the effect of probucol and losartan on preventing restenosis after balloon angioplasty in hypercholesterolaemic rabbits and the expression of growth factors. Methods 30 male New Zealand White rabbits were fed with hypercholesterolaemic diet. After one week of diet, all rabbits were balloon injured on iliac artery, then they were randomly divided into control group(group C), probucol group (group P, 1000mg·d-1) and losartan group (group L,10mg·kg-1·d-1). At the 1st, 8th , 21st and 35th day, The plasma concentrations of lipids were examined in all rabbits. At the end of 4 weeks after injury, the rabbits were killed in groups. The iliac arteries were examined with histomorphometrical method, examined Insulin-like Growth Factor-I Receptor(IGF-IR), Vascular Endothelial Growth Factor (VEGF) and Proliferating Cell Nuclear Antigen (PCNA) with immunohistochemical method. Results The plasma concentrations of lipids at the 8th day was higher than that at the 1st day in each group, which at the 21st day was higher than that at 8th day. But there was no statistical significant difference between that at the 35th day and that at the 21st day. The blood cholesterolaemic level had no statistical significant difference among all groups. Compared with that of group C, the lumen area, the intimal and medial area were larger in group P and group L, but there was no statistical significant difference in external elastic membrane (EEM) between two groups. There was no statistical significant difference between group P and group L. 55% of the increase in lumen area (+0.62mm2) was due to an increase in EEM (+0.34mm2) in group P, 65% of which (+0.54mm2) was due to an increase in intimal and medial area (-0.35mm2) in group L. The change of Lumen area correlated more strongly with the change of EEM(r=0.66,P<0.01)than with the change of intimal and medial area(r=0.38,P<0.01)in group P, which in group L correlated more strongly with the change of intimal and medial area(r=0.74,P<0.01)than with the change of EEM(r=0.28,P<0.01). Compared with that of group C, the expression of IGF-IR, VEGF and PCNA were decreased significantly in group P and group L. There was no statistical significant difference between them. Conclusion Probucol and losartan could prevent the restenosis of rabbit's iliac artery after balloon injury, decreasing the expression of IGF-IR, VEGF and PCNA. There was no statistical significant difference between two drugs.
