Object: To establish a rat model of Lishirezheng (syndrome of interior-excess-heat of TCM) associated with hypoxemia. Methods: This experiment consisted of two parts. Experiment one was to determine the length of ischemia time and reperfusion time that Lishirezheng could be formed. Experiment two was on the basis of the determination of intestine ischemia time. Ileic acid was injected through jugular vein 15 minutes before intestinal ischemia-reperfusion, the dosage was 0.05ml/kg, 0.075ml/kg and O.lOml/kg respectively. The mortality rate of each group was observed and 0.075ml/kg selected as the dosage of making models. Experiment three : The rat model of Lishirezheng associated with hypoxemia was made on basis of the first two experiments. Results: Experiment one: The 30-minute ischemia group had no differences as compared to sham operated controls. The belly perimeter and rectum temperature of the 45-minute ischemia group were higher than those of the sham operated controls, The wet weight of faeces and the wet-weight to dry-weight ratios were lower than controls after 12 hours, 24 hours and 48 hours of reperfusion(P<0.001).But the arterial gas analysis show no differences from sham operated controls. The belly perimeter and the wet-weight to dry-weight ratios of the 60-minute ischemia group were higher than controls, but the rectum temperature was lower, and some rats of the 60-minute group ischemia showed melena. Experiment two: The rats of control group and experimental I group had no death, The mortality rate of experimental II group was 30 percent, that of experimentallllgroup 80 percent. So the experimental II group was selected as the model group. The belly perimeter, rectum temperature, wet weight of faeces andwet-weight to dry-weight ratios after 12 hours, 24 hours and 48 hours of reperfusion had the same changes as experiment one. PaOj of the model group was decreased and AaDCh of the model group was increased obviously as compared to control. The manifestations of Lishirezheng could not be induced by injected ileic acid only. Conclusion: The rat model of Lishirezheng associated with hypoxemia could be established by injecting ileic acid through jugular vein before intestinal ischemia-reperfusion. The model had the following characteristics: stable, fair mortality rate, convenient and economy, thus providing a foundation for the study of therapeutic effects of certain herbs.
