Aim: To identify Guangxi Rongshui Paragonimus using molecular technique and study theirs gene variation and phylogenetic relationship s among relative populations. Method: extract the two worms DNA, design the especial primers using PCRdesn software to PCR, obtain the rRNA ITS2 gene sequence, then compare the gene identical with Blast program of GenBank, compare the gene variation with Primer Premier software, set up the phylogenetic tree using NEIGHBOR programs of PHYLIP software. ResuIt: gene identical comparison demonstrate: the ITS2 gene identical between Rongshui big and small cyst worms was identities=100% (389/389) ,Score=748 bits ; the ITS2 gene identical among Rongshui big cyst worm and Paragonimus skrjabini > Paragonimus macrorchis were Identities=100%(334/334), Score=662 bits; Identities=91%(308/336), Score=436bits, respectively; the ITS2 gene identical among Rongshui small cyst worm and Paragonimus skrjabini -, Paragonimus macrorchis were Identities=99%(360/361), Score=710 bits; Identities=92%(334/363), Score=484 bits , respectively ;gene variation comparison indicate: the genevariation number is 0 among big cyst worm> small cyst worm and Paragonimus skrjabini, the gene variation number are 0, 28, 26, 28, 33. 35, 25, 20, 28 following by Paragonimus miyazakii, P.ohirai, P.harinasutai, P.kellicotti* P.mexicanus ^ P.siamensis, P.westermani, P.heterotremus. P.macrorchis ;Phylogenetic tree indicate: Rongshui small cyst worm, big cyst worm and Paragonimus skrjabini lie at the same position of the phylogenetic tree , but great further with Paragonimus macrorchis. Compare with other species, Paragonimus miyazakii is closer with Paragonimus skrjabini in the phylogenetic relationships. Cone I us i on: according to the gene identical and classification criterion, suggest Rongshui small cyst worm> big cyst worm and Paragonimus skrjabini are high identical, big cyst worm and small cyst worm are identical with Paragonimus macrorchis; according the gene variation comparison, indicate Rongshui small cyst worm, big cyst worm and Paragonimus skrjabini have no obvious gene variation, but obvious gene variation with Paragonimus macrorchis; according the phylogenetic tree, indicate Rongshui small cyst worm ^ big cyst worm and Paragonimus skrjabini have no obvious phylogenetic distance , but have further distance with Paragonimus macrorchis, Paragonimus miyazakii is closer with Paragonimus skrjabini in phylogenetic relationships than other species. According the results of gene identical comparison -, gene variation comparison and phylogenetic tree, indicate Rongshui small cystworm, big cyst worm and Paragonimus skrjabini are the same species, small cyst worm is not Paragonimus macrorchis.
