Objectivesl.To establish the models of the acute rejection in small boweltransplantation in rats.2.To study the expression of the IL-2 and TNF-a in blood serum in different periods of small bowel transplantation, and the relationship between the IL-2 and TNF-a and the acute rejection.3. To analyze the changes of the selectin' s expression onprotein and gene level during small bowel allograft rejection in rats,and their significance in small bowel allograft rejection in rats. MethodsHeterotopic small bowel transplantation was performed with inbred rat F344/N and Wistar.All recipients were divided into four groups;group I , Wistar; group H , F344/N -*?F344/N;group III, F344/N -* Wistar/A;group IV , F344/N 棔 Wistar/A+FK506.Six rats were put to death on POD 3,5 and 7,and the intestine was cut under sterile condition, The serum was taken in vena cava.All samples were examined histologically and the expression of selectin were detected with immunohistochemistry-technique.The intra- graft mRNA expression of selectin were determined .The serum levels of TNF-a and IL-2 were measured with radioimmunoassay. Resultsl.The histological examinination showed that mild acute rejetion occurred-5-on POD 3 in groupIII,moderate acute rejection on POD 5,severe acute rejection on POD 7,while in group II ,no acute rejection was found .The histologic evidence of group IV indicated that FK506 could effectively control acute rejection.2. The serum levels of TNF-a and IL-2 measured with radioimmunoassay were highly elevated in group III on POD 3,significantly higher than that of other groups(P<0.01),and they were elevated higher with the aggravation of the acute rejection.While the serum levels of TNF-a and IL-2 measured with radioimmunoassay were a bit elevated in group II and group IV on POD 3 and POD 5,and then the tendency would descend,there was no significant difference between group II and group IV(P >0.05)3. selectin-P,selectin-E and selectin-L were highly elevated in group III on POD 3,significantly higher than that of other groups(P<0.01),and selectin-E and selectin-L were elevated higher with the aggravation of the acute rejection, while P-selectin was not significantly elevated. But the expression of the selectin was a bit elevated in group II and group IV after the operation , though the expression in group FV was lower than that in group II.Conclusions1. The serum levels of TNF-a and IL-2 measured with radioimmunoassay may closely correlate with small bowel transplantation rejection.we think that determining dynamically the levels of TNF-a and IL-2 can predict the occurrence of acute rejection at an early stage.2. Early expression of P-selectin and high expression persistent of E-selectin and L-selectin in small bowel transplantation rejection can predict the occurrence of acute rejection at an early stage.-6-3. The measurement of the mRNA content in selectin with RT-PCR can precisely show the seriousness of acute rejection.4. The expression of selectin and the tropism of white cells that is inter-linked by the selectin are at the early stage of acute rejection in small bowel transplantation , which will further cause the deterioration of acute rejection .So ,the measurement of the expression of all the selectin in mucosa issignificant to the early diagnosis and therapy of acute rejection.
