In order to detect the effect of human sperm mannose-ligand receptor on the fertilization ability, in the study reported here mannose-ligand receptors(MRs) were purified from human sperm by modified mannose-agarose gel affinity chromatography coloumn and determined protien concentration by Lowry, Preincubated zona-free hamster oocytes with four purified mannose-ligand receptor (pMR) concentrations before sperm penetration assay(SPA) to test the pMRs cell biology nature of inhibition to fertilization. Then preincubated hemizona pellucida with the known concentration pMRs (2.84u g/ml) , to study the effect of sperm binding to the zona on the hemizona assay(HZA).The result indicated: (1) 19.6?7.Ing pMR could be purified from 1 X 106 human sperm, its molecular weight is 67.8 ?1.6KDa.(2) Fertilization was inhibited partially when preincubated zona-free hamster oocytes with 0.95 u g/ml pMR, When preincubated them with 2.84 u g/ml pMR, fertization was absolutely inhibited (p<0.05) .(3) While preincubated human hemizona pellucida with 2.84 u g/ml pMR did not affect the binding of sperm to the hemizona pellucida(t=1.25,p<0.05). This result may indicate that the effect of pMRs on fertilization ability is not in sperm-zona pellucida recognition, but in sperm-egg fusion.
