Reactive oxygen species (ROS) and its lipid peroxidation have been proposed to be a major mechanism involved in lead intoxication. Tea polyphenols (TP), an effective antioxidant, has the function of ROS scavenging, inhibiting lipid peroxidation, reactivating antioxidant enzymes, chelating metal ions. The study was designed to probe into the effect of TP on lead induced lipid peroxidation.60 male Wistar rats were equally divided into six groups: the control group, Pb, Pb+TP50mg/kg, Pb+TPlOOmg/kg, Pb+TP300mg/kg, TPlOOmg/kg. At the beginning of the experiment, 50, 100, 300mg/kg body weight TP were given to rats orally , on the 6th ,8th , 10th day , 40mg/kg bwt lead acetate was given to rats by intra-peritoneal( ip ) method ,the control group were given distilled water orally and saline by ip , on the 11th day , blood from vein of orbit was used to make an experiment, and then all rats were raised as above till another week ,the rats were killed and the blood from arteria , veinous femoralis, and tissue of liver, brain were used to make an experiment.Malonaldehyde (MDA) , superoxide demutase (SOD) , glutathione (GSH) , gluthione-S-transferase (GST) were measured by the method of reagent' s directions .Statistical analysis of the data was performed by either single factor analysis of variance( ANOVA ) or student' s t-test using SAS for windows 6.12 statistical package, all values were expressed by the way of mean ?standard deviation.The results show as follows:1: There is a significant difference between the body weight of the lead-treated and that of the control group (PO.05), TP plays a great role in anti-body weight loss of rats3treated with lead .after 24 hours, 1 week of the last lead - treated , there was significant positive correlation between body weight of lead-treated rats and TP consuming ,the correlation is r = 0.973 ( P <0.001) , r = 0.9979 ( P O.0001) respectively .At the same time , TP can improve the lead-treated rats' general conditions .2: After 24 hours, 1 week of the last lead-treated, MDA of lead-treated rats was significantly higher ( P <0.05) and SOD activity was significantly inhibited ( P <0.01 ), as compared to the control group respectively , but TP100, 300mg/kg in use have the protective roles from lead poisonous effects . Both lead and TP have the function to increase GSH and GST in blood significantly but they have opposite effects on rats.3: After 1 week of the last lead treatment, in liver, there is a significant difference between MDA in lead-treated rats and the control group ( P <0.05), TPlOOmg/kg, TP300mg/kg in use can significantly protect animals' MDA from rise respectively (P O.05, P<0.05), and have a dosage -dependent effect . And at the same time, Pb has the effects of GSH deleption (PO.05), inhibiting SOD activity (P<0.05), inhibiting GST activity (P <0.01), TPlOOmg/kg, TP300mg/kg in use can significantly protect rats from GSH deleption ( P <0.05), SOD ( P <0 05) and GST ( P <0.05) activity inhibition respectively, and have dosage-dependent effects too.4: After 1 week of the last lead treatment, in brain ,there is a significant difference between MDA in the lead-treated rats and that of the control group (P <0.05), TP300mg/kg in use can significantly protect animals' brain MDA from rise ( P <0.05), and have a dosage-dependent effect. At the same time, Pb has the effects of GSH deleption (P <0.05), inhibiting SOD activity ( P <0.01), inducing GST activity ( P <0.01), TPlOOmg/kg, TP300mg/kg in use can significantly protect rats from GSH deleption (P <0.05), SOD (P <0.05) activity inhibiting and GST activity rising( P <0.05) respectively, TP50mg/kg,TP100mg/kg,TP300mg/kg can significantly prevent SOD activity from decreasing ( P <0.05).In conclusion, lead can induce lipid peroxidation in the blood x liver?brain of rats, howerer ,TP 100, 300mg/kg orally used may influence antioxidant enzymes and the level of GSH to protect rats from lead-induced lipid peroxidation , protect rats from lead inhibition4to growth, body weight loss...
