Objective:Observe the effect of Shenfu injection on withdrawal syndromes and neurotransmitter of morphine-abstinent mice and on intracellular calcium level in the morphine-dependent neuronal cells line. To study the detoxification of Shenfu injection on morphine-dependent mice and expore the mechanism of its effect.Methods:Morphine-dependent mice established by injecting of morphine on dosage increasing by degrees. The effect of Shenfu injection on the abstinent syndromes, including body weight were studied .The hypothalamic monoamine neurotransmitters such as NA, DA, 5-HT were tested by fluorospectrophotometry. Morphine-dependent neuronal cells were grown in RPMI164O medium(containing lO5mol morphine).The effect of Shenfu injection on intracellular calcium level was observed by Fluo-3-AM measurements using Flow Cytometry (FCM) in the morphine-dependent human neuroblastemar SK-N-SH cells line.Results:Shenfu injection could alleviate withdrawal syndromes, recover the the loss of body weights and decrease the conentration ~of the neurotransmitters. Intracellular calcium level in morphine-dependent cells were remarkably higher than control ones. During naloxone-precipitated3withdrawal syndrome elevated Ca2~ levels quickly reduce. Shenfu injection could prevent this effect ,maitain the calcium levels.Conclusion: (1) Shenfu injection has some effects on morphine梔epedent mice which may relate to inhibiting neurotransmitters. (2) The?effect of Shenfu injection on tolerant nerve cells makes known that its detoxification mechanism has relationship with regulating the calcium level in the morphine梔ependent cells.
