| Objective: To observe and understand the secretion and expression of IL-8 and the infiltration and accumulation of polyrnorphonuclear leukocytes (PMNL) in the injuried brain at different phases. IL-8.. PM7NL and histopathology were also studied . We also to evaluate the comtributions of IL-8 and PMNL to the secondary brain injury(SBI). Methods: Using the TBI models , the damaged region of brain was removed in different phases and stained with HE and immunohistochemixal to evaluate the histopathologic changes . The changes of PMNL aggregation infiltration and expression of IL-8 in brain and correlations among PMNL IL-8 and histopathology at different phases after cerebral injury was also studied. Results: There are not expression of IL-8 and PMNL infiltration in normal brain. PMNL aggregation and infiltration and postive expression of IL-8 in brain progressively increased following histopathologic exacebation. Expression of IL-8 and PMNL infiltration can be only found in injuried cortex, contralateral cortex and bilateral hippocampus can抰 be seen. The postive expression of LL-8 was in the vascular wall, infiltrated PMNL and astrocytes. It was firstly seen at 2 hours after trauma , while PMNL infiltration was firstly seen at 6 hours. Both peaked at 24 hours after TBI, and marked decreased after 48 hours. 3 IL-S ~A4J PMNL ~ ~JT~t T~ ~ $4~ Conclusions: 0 A local inflammatory response was initiated immediately following TBI and is characterized by the infiltration and acumulation of PMNL. And the inflammation may play an important role in SBL ㊣L-8 promotes invasion and accumulation of PMNL into brain parenchyma 甇ur result indicated that there in a postive relationship between PMNL and IL-8 and they are intimately connected with the histopathology of the brain.Furthermore, with the histopathology of the brain , expression of IL-8 is earlier than PMINL infiltration OWe demonstrated that brain endothelial cells infiltrated PM7NL and astrocyte mainly produce and secrete IL-8.
