We have developed microcapsules using sodium (SA) and Poly-L-Lysine (PLL). APA microcapsules have proven effective in protecting enclosed live cells from immune rejection following transplantation into experimental animals, thereby eliminating the need for immunosuppressive therapy. The development of the electrostatic droplet generator result in considerably smaller capsules (150-3 OOum in diameter) with improved sphericity, surface smoothness and, most importantly, capsule strength. A factorial design method of screening was chosen to study influences of different experimental parameters on size and stability of capsules.A study of 6 parameters [electrostatic pulse, pump flow rate, needle gauge, alginate viscosity, PLL concentration and PLL reaction time] showed optimal condition. By utilizing laser scanning confocal microscopy and FITC labeled dextrans with different molecular weight, it was found that the microcapsule-6-membrane could cut off dextrans with MW beyond 11 OkDa. The mechanical strength of different types of microcapsules was measured by mechanical shaking experiments. The results showed that APA microcapsule with high mechanical strength was due to the membrane formed with alginate and polylysine.The experiment result of empty APA and APA-BCCs implanted into the rats indicated that APA microcapsules were biocompatible.The hemiparkinsonian symptoms of PD monkeys grafted APA-BCCs have been improved after 24 months. The transplantation of microencapsulated BCCs can increase the content of dopamine and metabolites in PD monkeys. APA-BCCs had more significant effects than BCC, this demonstrates APA microcapsules have markedly immuno-isolated effects.From the above results, we can draw that APA micro-capsule can cut off immune cells and function as efficient immunoisolating barrier.
