ABSTRACT Objective: Graft-versus-host-disease (GVI-LD) and graft-versus-leukemia (GVL), which accompany allogeneic hematopoitic stem cell transplantation (allo-IISCT) for leukemia, could significantly affect therapeutic effect after allo-HSCT. Now, it is well known that T cells play a central role in GVI-ID and GVL. identifying specific GVHD and GVL T cell to do selective donor T-cell-depletion(TCD) transplantation can improve the effect of leukemia in allo-HSCT. investigation of the distribution of TCR V P subfamily T clonal cells in patients following allo-HSCT and the research of the acts of T cells in GVHD and GVL can provide valuable materials for selective lCD allo-HSCT and T cell adoptive immunotherapy. Methods: In order to research the distribution and clonality of TCR VP subfamily T cells before and after allo-HSCT, complementarily determining region 3(CDR3) size of I cell antigen receptor(TCR) 24 variable region gene was analyzed in peripheral blood lymphocytes(PBLs) drawn from 8 allo-HSCT cases with the use of reverse transcriptase-polymerase chain reaction(RT-PCR) and genescan analysis. Results: Data obtained from 10 normal individuals indicated that all V P subfaniilies of T cells were polyclonal expressed while skewed expression of the V P repertoire from 2 compatriot donors in PBLs. None or only few V P subfamily T cells could be found in patients of pre-transplantation and the early of post-transplantation, and individual otherness was the general characteristic of dominant TCR V P subfamily T cells. New T clonal cells could be found as the time passed following transplantation. Skewing distribution and oligoclonal expansion of ICR V P subfamily T cells could be found in each patient with GVHD after -3- allo-HSCT. Conclusioii: (1) Clonal expansion and dominance are the character of ICR V P T cells in allo-HSCT patients. (2) Representation of oligoclonal expansion of TCR V P subfamily T cells can be related with GVHD and GVL activity.
