Reserch On The Relationship Of Streptococcal Preparation Antitumor Effect And Cytokines Secretion

Objective: (1 )To observe the relationship between preparation concentration, inducing time and jteS antitumor effect and cytokine secretion.(2)To study the antitumor mechanism of domestic streptococcal preparation Sapeilin from the level of cellular and cytokine. Methods: Using different concentration of streptococcal preparation to culture with the peripheral blood lymphocyte(PBL) of normal persons and tumor patients, separately. When PBL was cultured in different times (24, 48, 72 hrs) then the PBL effect on K562 cell and Raji cell was detected by MTT method, and the levels of IL- 12, IL-2, IFN- ~, TNF- a in PBL cultural medium were evaluated either by the methods of ELISA or biological activity. Results: PBL had different effect on inhibition K562 cell and Raji cell after induced by different concentration Sapeilin for different times and the levels of cytokines secreted by PBL were also different. (1)0. iKE/mI and 0.O2KE/ml Sapeilin could enhance the ability of PBL to inhibit tumor ceiis,especially, the 0.O2KE/ml concentration is much better;And the high levels of IL-12, IL-2, IFN- i? TNF- a could be detected in it's cultural medium. (2)The levels of IL- 12 and TNF- a were highest in 24hr,and the secretion of 1L-2 and IFN- a was much more in 72hr than that of in 24hr.(3)There was no significant differences of antitumor effect and cytokine secretion for PBL between the group induced by 0.OO4KE/ml and the control group.(4)O5KE/ml Sapeilin inhibited not only PBL ability of antitumor effect, but also its cytokine secretion. The effect on inhibition K562 by PBL induced from Sapeilin was stronger than that of Raji cells which was NK resisted. Tumor patients?immune condition had influence on PBL induced by Sapeilin. Conclusions: After Sapeilin inducing, PBL of normal persons and tumor patients effect on inhibition tumor cells was related with Sapeilin concentration and the inducing time.
