Abstract/Summary:
Among neuroglial cells, astrocytes are the most in number and distrbution. Astrocytes take on most functions of neuroglial cells?s, such as supporting, repairing and regeneration, they play important roles in physiology and pathology of nervous system. According to the theory of astrocytes lineages, astrocytes are divide into two types: type 1 astrocytes(T1A) and type 2 astrocytes(T2A). Objective: To establish the method of purification culture of TIA and T2A, to study the biological characters of T1A and T2A, and to build a solid foundation for the subsequentant reasearch. Methods: Using shake culture and differential adhesion, T1A and T2A were separated and purified; with the immunocytochemical method, the expressions of PP in TLA and T2A were observed; treating PCL2 cell with the medium conditioned by hA or T2A, the outgrowth of neurites of PCI2 cells were observed, the viability of PC12 cells were investigated with the method of MIT. Result: (1) With shake culture and differential adhesion, TIA and 0?A progenitor cells were seperated, and purified TIA were obtained; (2) By differential adhesion, 0?A progenitor cells were purified, which A2B5 reactivity is positive; (3) Induced by 10?0% serum , 0?A progenitor cells developed into T2A; (4)TLA and T2A were different in morphology. (.5) TLA expressed PP and T2A did not; (6) Comparing with the control group, PCI2 cells treated with the TLA or T2A conditioned medium had significant outgrowth of neurites. And the value ofOD had arised from 0.278 to 0.512(p<0. 001) and 0. 566(pKO. 000. Conclusion: (1) We have established the method of purification culture of I LA and T2A, which is suitable for our lab. (2)T1A and T2A are different in morphology. (3) T1A expresses-APP and T2A does not, implying that TIA play an important role in the pathogenic pathway in AD. (4) T1A or T2A conditioned medium can promote PCIE2 cell?s growth, suggesting that TIA and T2A are helpful to neural regeneration. (5) Studies on 0?A progenitor cells, T1A and 12A are important to the treatment of regressive diseases in central nervous system.
