Objective Tripterygium wilfordii can relieve the airway hyper-responsiveness and inhabit the inflammatory response in the asthma model, meanwhile it can also regulate expressions of cytokines which participate in asthma. It is still unknown the effects of Tripterygium wilfordii inhalation on asthma for in most studies it was injected intraperitoneally in most animal experiments. In this study the effect of tripolide inhalation on asthma in ovalbumin-sensitized guinea pig was, and how it affect the properties of the K channels of airway smooth muscles of guinea-pig trachea. Methods 60 guinea pigs weiting about 250-300g were divided into 5 groups. The animals were sensitized with ovalbumin suspension according to the previous study. In the study day , tripolide (8mg/mi) was inhalated for 30 minute in the tripolide group, dexamethasone was injected intraperitoneally in the dexamethasone group, and the two medicine were both given in the tripolide-dexamethasone therapy group before the ovalbumin was inhaled. Another two 12 guinea pigs, sensitized and non-sensitized as asthmatic control group and normal control group. Seven days later airway responsiveness was measured with PC2O; morphological examination was taken by light- and electron-microscope. The area and percentage of bronchiole tube wall were examined by Image Analysis system. Properties of the K+ channels of airway smooth muscles in guinea-pig trachea of normal control group, asthmatic control group and tripolide group were recorded.A1l the data were statistically analysed with Lasmaster DME system and PCLAMP 6.02 software. Results (1) PC2O in asthmatic control group (4.333E-2 ? .966E-2) is much higher than in normal control group (0.3467 ?0.1573) . PC2O in tripolide group (0.1333 ?4.131 E-2) is less than in asthmatic control group, but there was no significant difference between them(P>0.05). PC20 in dexamethasone group (0.2000 ?9.798E-2) is much less than in asthmatic control group and tripolide group (P<0.01) . There is no difference between that in tripolide- dexamethasone group ( 0.1333 ?4.131E-2 ) and normal control group(P0.3 16, >0.05). (2) The area and percentage of bronchiole tube wall in asthmatic control group (58597.84 ?23020.87; 0.8570 ?4.27E-2) were much higher than that in normal control group (30626.51 ?14085.43; 0.5764 ?6.60E-2) (P<0.01)0 In tripolide group (40641.36?15398.66; 0.6929?.126) dexamethasone group (33721.64?8556.53; 0.6884?.68E-2) , and tripolide-dexamethasone group (26981.68 ?7748.35; 0.5996 ?7.82E-2) they decreased gradually (P<0.01) . Those in tripolide-dexamethasone group reduced to normal (P=0.631, >0.05; P0.465, >0.05) (3) In model group the fimdimembrane of capillary vessel endotheliocyte and pulmonary vesicle epithelial cell grew much thicker; trophonemas of type II pulmonary vesicle epithelial cells disorganized, even necrosis or ecclasis partly; tight-injunction relaxed, electron density debased and become clouding; acidophil granules cells infiltrate into septum and cavern of pulmonary vesicle. After tripolide was administered the thickness of fimdi-membrane decreased; trophonemas of type II pulmonary vesicle epithelial cells which had been injured were partly recovered, but still tenuous; tight-junction got stronger and there were also cidophil granules cells infiltrate but the number reduced greatly.
