Artificial insemination technique can improve the pig economic efficiency,generalize the good kinds of boar semen, and accelerate the process of breed improvement. The level of preservation of boar semen, is the primary links that improve artificial insemination techniques, and greatly improve the boar semen used in time and area. Storage of boar semen at room temperature has been widely used in artificial insemination,it improves the utilization efficiency of boars, and generalizing artificial insemination techniques has great significance. In order to effectively carry out artificial insemination technology, pig semen dilution preservation is the key technology.This study used two kinds of formulations, designed 10 swine semen diluted formula for boar semen stored at room temperature. Results that tested sperm motility after semen storage showed that : powder I, Powder II, I and II, sperm viability, overall survival at all times was significantly higher than (p <0.01) III, IV, powder III, powder IV, and were significantly higher than (p <0.05) V, powder V, and V, powder V were significantly higher than (p <0.05) III, IV, powder III, powder IV; Although the powder I, powder II, these three indicators were slightly higher than the I, II, but the difference was not significant (p> 0.05).It obtains a relatively good preservation effect.Screening out four kinds of dilution I, II, Powder I, Powder II is as the best formula; semen preserved in the formula for 3 days and 4.5 ~ 5 days, and the live rates were 0.5 and 0.3.In order to effectively verify the preservation effect of the best dilution, ensure that after dilution and stored, semen has a high fertility, it used aniline blue - crystal violet staining of the acrosome of pig sperm stored at room temperature, the rate of acrosome intactness, when diluted and stored for 72h and for 120h,is respectively, 95% and 70% or more. Studying that temperature and pH value on the effect of semen stored at room temperature, results showed that: the best storage temperature is 15℃~ 20℃; the best pH value is 6.5 ~ 6.9.To test the effect of diluting the application of liquid formulations, it used Powder I, Powder II, I and II formula for semen dilution, results that artificial insemination of sows tested show that: Powder I, Powder II, I and II, formula diluted preservation of semen pregnancy rate and the average litter size was significantly higher than (p <0.05) of natural mating pregnancy rate and the average litter size. It indicated that Powder I, Powder II, I and II, formula diluted stored semen achieve an ideal result in artificial insemination of fertilization .the powder diluent powderⅠ, powderⅡ, not only have good results, strong stability, and transport and carry easily, but also production process is simple, as a product-type preservation of boar semen diluted solution, it is more suitable for pig production in the popularization and application. In this study, diluted solution stored at room temperature in pig production and practices provides a frame of reference.This research developed semen diluent powder, diluted, and expanded the capacity of semen, extended semen effective storage time, and icompared with natural mating, it improves conception rate and litter size of sows. In the large-scale pig farm applications, it may significantly increase the economic benefits of pig.
