| In this study,the endophic bacteria were seperated and selected fristly from the stem of Pinus massoniana and Pinus thunbergii,and the community population of endophytic bacteria surveyed systematically. Through surface disinfection and antibiotic marker techniques,seperating the endophic bacteria from Pinus massoniana and Pinus thunbergii and with the routine bacteria tests,Biolog and 16S rDNA analysis to identify them. Then through dual-culture tests to screen the antagonistic microbia. This study supplies the basic theory to explore the relationship between endogenous and antagonistic characters of endophytic bacteria. The results as follows:1. Seperating the endophytic bacteria from the stem of Pinus massoniana and Pinus thunbergii during the whole year:thirty-nine strains were seperated from them. Out of thirty-nine strains, twenty strains were seperated from Pinus massoniana,ninteen strains were seperated from Pinus thunbergii. The population dynamic vartion not change sharply during the year.2. Through seed soaking and pot experiments,nine strains out of thirty-nine bacteria were retrieved and justified as the endophic bacteria. They are NSM-03, NSM-04, NSM-05, NSM-07,NSM-12,NSM-17,NSM-19,NSH-07 and NSH-18.3. With the the observation of figuration characters,the phsiological and biochmeical tests, combined with Biolog and 16S rDNA gene analysis, those nine endophic bacteria are identified as Bacillus megaterium, Bacillus amyloliquefaciens,Burkholderia cenocepacia, Agrobacterium rubi, Curtobaterium flaccumfaciens, Enterbacter cloacae, Enterobacter cloacae,Enterbacter agglomerans and Acinetobacter calcoaceticus.4. Through dual-culture tests, NSM-05 has the antagonistic effection on all fifteen kinds of pathogenic bacteria.NSM-17 has the antagonistic effection on one pathogenic bacterium and NSM-19 has the antagonistic effection on two out of fifteen pathogenic bacteria. |
