The genetic diversity and kinship of different varieties of Camellia oleifera and Paulowniawere studied by ISSR marker. And varieties of Camellia oleifera and Paulownia were identifiedby both molecular and micro-morphological markers. The main results were shown as follows:1. The genetic diversity of different varieties of Camellia oleifera was high. A total of 6 ISSRPrimers were employed on 66 individuals of cultivated Camellia oleifera from 100 Primers. Anda total of 121 bands were amplified with 112 polymorphic bands and percentage of polymorphicbands(PPB) was 92.56%. The average value of observed number of alleles (A), average value ofeffective number of alleles (Ne), Nei's gene diversity (He) and Shannon's information index (I)were 1.9339,1.6509, 0.3698,0.5417 respectively. The size of band amplified was between 100bpand 1200bp.2. Paulownia species possessed low genetic diversity. A total of 5 ISSR Primers wereemployed on 10 individuals of cultivated Paulownia from 100 Primers. And a total of 46 bandswere amplified with 18 polymorphic bands and percentage of polymorphic bands(PPB) was39.13%. The average value of observed number of alleles (A), average value of effective numberof alleles (Ne), Nei's gene diversity (He) and Shannon's information index (I) were 0.4934,0.3418,0.1870,0.2715 respectively. The size of band amplified was between 250bp and 1000bp.3. According to the ISSR results,the coefficients of genetic similarity in different Camelliaoleifera and Paulownia cultivars were studied. Cluster analysis dendrograms of both Camelliaoleifera and Paulownia cultivars were drawn. Genetic diversity and similarity as well as kinshipof different Camellia oleifera and Paulownia cultivars were revealed. Genetic distance indifferent Camellia oleifera and Paulownia cultivars were further determined.4. The application of molecular markers in variety indentification of Camellia oleifera wasalso investigated. And the varieties of Paulownia were identified by using micro-structure ofsprig cross-section and epidermis hairs.
