Quercus acutissima is widely distributed in subtropical and temperate regions of China. It is applied widely and also has vast potential developing. Asexual propagation of Quercus acutissima was difficulty. It has great significance of tissue culture and cutting propagation. In this research, tissue culture and cutting propagation were studied and the main results were as follows:1.The tissue culture technology of Quercus acutissima(1) The best explants were the seedlings of seed germination and sprout of trees.(2) The results of sterilization of explant showed that the lowest rate of pollution after sterilization was 16.5% and the rate of germination was 73.1% by using explants from sprout tree. The lowest rate of pollution was 17.5% and the rate of germination was 82% by using explants of seedlings stem from seed germination.(3) The best medium for the bud induction was MS+6-BA1.0 mg·L-1 and the germination rate was 83.3%.(4) The best medium for the bud proliferation was MS+6-BA1.0 mg·L-1+NAA0.02 mg·L-1 and the proliferation rate was 4.6.(5) The best medium for rooting was 1/2MS + NAA0.5mg·L-1 and the rooting rate was about 87%. The survival rate of transplanting was 80%.(6)The results of callus induction showed that the best medium was MS+2,4-D1.5mg·L-1 .2. The techniques of cutting and seedling of Quercus acutissima(1) The survival rate of middle part of the cutting was slightly higher than the tip and the base of cuttings.(2) The result of nine kinds of source showed that the highest survival rate was 27.1% of Chizhou provenance in Anhui.(3) One year old seedlings completely lignification was used in hardwood cuttings. It germinated of one year old seedlings after stumping. The survival rate was 40% after soaked two hours by concentration of 200ppm of ABT-1.(4) The best effect of softwood cuttings was on June 20th. The survival rate of one year old seedlings was 56.7% after soaked two hours by concentration of 200ppm of ABT-1.(5) The cutting survival rate of sawtooth oak was positive correlation with iPA,IAA,ZR,and GA . It was negative correlation with the concentration of ABA. The higher concentration of ABA may was an important reason for the low survival rate.(6) The cuttings survival rate of stump plant of seedlings was higher than the trees sprout both in softwood and hardwood cuttings. This may be related to the higher ratio of N and P and the lower ratio of C and N.
