China is the world's largest rice production country in the world.While the influences of environmental stress are important factors in rice production.According to statistics, abiotic stress leading to a reduction of 50-80% in crop production.With the development of molecular biology techniques, many stress resistance genes have been cloned and overexpressed in rice by using Agrobacterium, in order to verify their function. The further reserch of these genes'function will help to reveal the molecular mechanism of stress resistance of rice, and it will provide us a theoretical basis on rice molecular breeding.The expression of HSP90 is induced by various enviromental stesses.The reserch of HSP90 is very important to the study of biological stress.We found that OsHSP90 could be induced by heat,cold,drought and high salt stress.It suggests that OsHSP90 may serve as a common regulatory factor in rice stress response. As to research the stress resistance mechanism of OsHSP90, we chosen OsHSP90 as bait protein,according yeast two-hybrid system.finally, we dicovered a number of OsHSP90 interactions proteins. we have chosen a rice PSII subunit like protein gene (OsPSâ…¡-Like) as our study object, expected to discover its physiological function, stress expression and regulation mechanism by molecular biology techniques.ultimately, then it will establish a basis on high photosynthetic efficiency cultivar selection and crop production.The results are as follows:1. According to the approaches of yeast two-hybrid system,we take OsHsp90 as bait protein,the interaction protein OsPSâ…¡-Like is filtered from the low tempreture and drought cDNA library of rice.2. Analyzing OsPSâ…¡-Like by comparing in the database,the results are,the full length of cDNA is 775bp,its coding area lies in 66-479bp encoding 137 amino acid residues.molecular weight is predicted to be 14.1KD,isoelectric point is about 9.86. N-terminal area has a nuclear localization signal KKAKK, we found that 92-120 amino acids sequence has an ADH_SHORT functional domains (Short-chain dehydrogenases/reductases family signature),40-137 amino acid sequence has a PsbR (Photosystemâ…¡10 kDa polypeptide) motif.3. Using the method of semi-quantitative RT-PCR to analyze OsPSâ…¡-Like and OsHSP90 expression at low temperature, high temperature, drought and high salt stress and in different physiological stage.Both of OsPSâ…¡-Like and OsHSP90 are found to express in different degrees before dealing with adversity stress. The result is that they are induced by adversity stress.they show differently expression pattern in different time and tissues at different physiological stages. 4. Constructing expression vector of sense,antisense cDNA fragment of OsPSâ…¡-Like and establishing the genetic transform system for rice mediatied by agrobacterium tumefaciens:direct sequence transform to the rice for overexpression is to amplify the physiological function of the gene, reverse sequence transfer is to prevented the expression of OsPSâ…¡-Like in rice by homologous sequences inhibition,consequently, to study the physiological function of the gene expression on rice resistant response.Meanwhile, we constructed a green fluorescent protein vector in order to determine subcellular localization of the OsPSâ…¡-Like to get futher study.
