| Florfenicol, as a new type amphenicols antibiotic and possessing well antibacterial effect and less untoward reaction, was to utilized extensively in prevention,control and therapy of bird species diseases. At the same time, people pay more attention to the question of florfenicol and it's metabolic products residue because of it's hazardous to food safety and destruction to natural environment. In this thesis, the degradable properties of florfenicol in hen dung was investigated at different environment. Based on its optimal degradation conditions, the reliable methods for excreta treatment and environment monitoring in economic manufacture.The chicken feces's florfenicol rudimental detection methods was established through adding factitiously florfenicol in hen dung and applying gas chromatography. The result indicate that no impurity waveform present near the waiting detection reagent waveform in the chromatogram, and have best specificity. The lowest detectable limit is 10μg/kg and the lowest limit of quantitation is 15μg/kg. A well linear relationship have displayed in concentration limit in 15μg/kg to 20,000μg/kg,and r=0.9991. When florfenicol concentration in hen dung is 15μg/kg,1,000μg/kg and 10,000μg/kg, coefficient of recovery touched 76-89%, the coefficient of variability in the same day was 1-2%, and in different days was 4-5%.The chicken feces treatmented by 20℃and protection from light, florfenicol degradation velocity display very fast, the half life period was 1.9 d, degradation kinetic equation was Ct=7.81×e-0.3699t, degradation rate constant was 0.3699,and degradation rate reach 90% approximately 11 d. florfenicol degradation capability descend strikingly when hen dung was handled with high temperature sterilizing and exorcizing organic matter. The half life period enhanced to 6.4 d and 7.2 d respectively, and degradation rate constant descend to 0.1083 and 0.0750 divisively. The different initial concentration of drugs addition quantity had disparate influence to the degradation of florfenicol. This influence have no evident change in light concentration, but when the initial concentration rise to 20 mg/kg, degradation has be inhibited. Also, the half life period,degradation kinetic equation and degradation rate constant of florfenicol degradation in hen dung hadn't be effected obviously by illumination.This research indicated that the main degradation style of florfenicol in chicken feces is microbial degradation, and the florfenicol degradation velocity step down obviously after removeling organic matter in hen dung. Exorbitant antibiotics initial concentration restrain Microorganisms growth and multiply then influence the degradation velocity. Florfenicol can't be degraded obviously by illumination.. |
