The study was carried out to determine the effect of deits with different zinc sources and different level Zinc Nicotinate on growth performance of growing pig, biochemical index , zinc enzymes in Serum, content of Zinc in serum ,content of Zinc swine feces , content and expression of Liver and kidney metallothionein in mice,while discuss the suitable zinc supplement of Zinc Nicotinate in diet and evaluated index of zinc nutritional status in mice.1 Effect of deits with different Zinc sources on growth performance of growing pig, biochemical index , zinc enzymes in Serum, content of Zinc in serum and swine feces105 growing pigs selected were divided into five groups,feeding trial was conducted at the feeding design was as follow: Control group:basal diet +120mg/kg Zn from ZnSO4 Trial groupâ… : basal diet +120mg/kg Zn from Zinc Nicotinate Trial groupâ…¡: basal diet +100 mg/kg Zn from Zinc Nicotinate Trial groupâ…¢:basal diet +80 mg/kg Zn from Zinc Nicotinate Trial groupâ…£: basal diet +100 mg/kg Zn from Zinc Methionine Each group had 3 repeat and each used 7 pigsThe results of feed trial indicated that: the ADG and ADFI of groupâ… ,â…¡,â…£were higher than that of Control group, whileâ…¢lower than Control group at the same time, the FCR of the groupâ… ,â…¡,â…£decreased at different, but the trial groupâ…¢raised slightly; The ALB content of the groupâ… ,â…¡,â…¢,â…£were lower than the control group but not significantly; The TP content of trial groupsâ… were slightly higher than control group while the other of trial groups were lower than control group ; The activity of AST and ALT of trial groups were not significantly different with control group ; The activity of AKP of the trial groupâ… ,â…£is higher than control group while the trial groupâ…¡,â…¢were lower than control group, and the activity of AKP of the trial groupâ…¢is significantly lower than other group; The activity of T-SOD of trial groups were higher than the control group, while the trial groupâ… were significantly different with control group and the trial groupâ…¢; The activity of Cu/ZnSOD of trial groups were higher than the control group, and the trial groupâ… were significantly different with the trial groupâ…£; The activity of LDH were lower than the control group but not significantly different. It indicated that the Effect of diets with zinc-rmethionine and different levels Zinc Nicotinate on growth performance was Concurrented with ZnSO4, The activity of AKP,T-SOD,CuZn-SOD of the trial groups were regularly different with the control group ,the variation of other biochemical parameters was not regular.2 Effect of deits with different Zinc sources on content of Zinc in serum and swine fecesTo determine zinc content in serum and swine feces , and the result showed the zinc content in feces diminished significantly with the reduction of dosage of zinc nicotinate, but the serum zinc level had no significant change compared to that of control group. It showed that zinc nicotinate in diets could increase body's zinc absorption and reduce degree of pollution.3 Effect of deits with different Zinc sources on zinc enzymes in Serum,content of Zinc in serum and swine feces and content of MTSixty mice were randomly divided into four treatment to investigate the effect of zinc sulfate and zinc methionine on growth and their possible regulating mechanism. feeding trial was conducted at the feeding design was as follow: Control group:basal diet Trial groupâ… : basal diet +30 mg/kg Zn from ZnSO4 Trial groupâ…¡: basal diet +15 mg/kg Zn from Zinc Nicotinate Trial groupâ…¢:basal diet +30 mg/kg Zn from Zinc Nicotinate Each group had 3 repeat and each used 5 miceThe results showed that: the diets with zinc could increase viscera index, while the Thymus and spleen index were significantly different; activity of AKP,T-SOD and CuZn-SOD, serum and tissues zinc content, liver and kidneys MT content were significantly increased by both forms and different levels of zinc, while trial groups markedly up-regulated activity of AKP,zinc content of liver and the level of liver MT.So activity of AKP,zinc content of serum and liver, level of liver MT could evaluate the zinc nutritional status in mice.4 Effect of deits with different Zinc sources on expression of metallothioneion-2Late in the third experiment, mice in control group and trial groupâ… ,â…¢were randomly elected to investigate content MT-2 mRNA of liver and kidneys, MT-2 mRNA were determined using semi-quantification reverse transcript polymerase chain reaction (RT-PCR). The results showed that the trial groups significantly increased MT-2 mRNA in liver. Also the two trial groups ,While MT-2 mRNA of kidneys abundance showed no difference between control and trial group. So expression of liver MT-2 mRNA could evaluate the zinc nutritional status in mice.According to above results, Zinc Nicotinate was a better additive, less dosage could play the similar role as more dosage of ZnSO4. Also it reduced the degree of pollution and diminished resource waste. And activity of AKP, zinc content of serum and liver, level of liver MT, expression of liver MT-2 mRNA could evaluate the zinc nutritional status in mice.
