This experiment was conducted to research the relationship between the formation and activation of osteoclasts and bone metabolism of the poultry in the cellular and molecular levels. On the basis of induce-cultured osteoclasts of chick embryo, the form and function of these OC were observed and identified by phase-contrast microscope, histochem ical staining for tartrate-resistant acid phosphatase (TRAP) staining at the first and the every other day, and detection of bone resorption pits through scanning electron microscopy at the seventh day. To study the impact on osteoclasts formation, activation and the function of bone resorption for the factors of calcium and phosphorus, and provide the scientific theory for researching the bone metabolism disease of poultry . The test results are as follows:(1) The osteoclats of the chick embryo were cultured by using 1,25-Dhydroxyvitamin D3 induction. Specificity of osteoclasts was identified by the means that mentioned above.The results proved that the induced osteoclasts had the activity of mature osteclasts,and the best induced concentration of 1,25-Dhydroxyvitamin D3 was 10-8mol/L;(2) Without the induction of 1,25-Dhydroxyvitamin D3, and add different concentrations of calcium, phosphorus to the test groups, we found that high concentrations of calcium, phosphorus could inhibit the of viability osteoclats in short time(<16h) , while middleconcentration groups and low concentration groups inhibited the of viability osteoclats 24 hours later . To 32 hours , the difference between high concentration groups and middle concentration groups was not significant, but lower concentrations were significantly different from middle concentration groups, and the difference between each test group and control group was significant;(3) The results of adding different concentrations of calcium and phosphorus to the osteoclasts cultured induce system showed: middle and high concentrations of calcium and phosphorus inhibited the the induction of 1,25-Dhydroxyvitamin D3 for the formation and bone resorption activity of osteoclasts;(4) We added different proportions of calcium to phosphorus to the system of osteoclasts induce-culture of the chick embryo in vitro to studied the impact on the osteoclasts formation, activation and function of bone resorption. The results showed that the inhibitory action was biggest when ratio of calcium to phosphours is 2:1, and the difference between the control group and test group was very significant.
