This thesis was conducted to study the effect of polysaccharides extracted from Allium Mongolicum Regel on activation of peritoneal macrophages and antioxygen in mice. The main contents and conclusions of paper were as follows:1. The effect of Polysaccharides extracted from Allium Mongolicum Regel levels (50 , 100 ,200 , 400 , 800μg/ml)on the phagocytosis capability of the mouse peritoneal macrophages,energy metabolism level and IL-6 were studied by neutral red phagocytosis test, MTT method and ELISA method. The results indicated that the concentration of Allium Mongolicum Regel Polysaccharides was 100μg/ml, which was the best concentration for activation of peritoneal macrophages.2. To determine the activation of 100μg/ml Allium Mongolicum Regel polysaccharides to peritoneal macrophages after 0, 12, 24, 36, 48h, was tested following the above methods. The results showed that the effect of polysaccharides extracted from Allium Mongolicum Regel on activation of peritoneal macrophages at 24h was better.3. O2-? and ?OH were cleaned up by polysaccharides extracted from Allium Mongolicum Regel in vitro antioxygen test. The results indicated that O2-?and ?OH were cleaned up by the polysaccharides extracted from Allium Mongolicum Regel. 5mg/ml of polysaccharide extracted from Allium Mongolicum Regel has the strongest ability to clean up O2-? ; And 1mg/ml of polysaccharide extracted from Allium Mongolicum Regel has the ability to inhibit ?OH strongly.4. In this experiment, 50 mices were randomly divided into 5 groups of 10mices. Blank control group was given 0.9% saline per day, positive control group was given 30 mg/kg ? BW VE, different doses of Allium Mongolicum Regel polysaccharide groups were 800, 400, 200 mg/kg ? BW, respectively. Preparatory experimental period was 7 days. After 15 days of the formal trial period, blood was sampled from eyeball and serum was separated, to investigated the antioxydation effects of polysaccharides extracted from Allium Mongolicum Regel by T-SOD, MDA, CAT, T-AOC, GSH-PX of serum, liver, kidney and heart in mice. The results indicated that polysaccharides extracted from Allium Mongolicum Regel has a certain antioxidant effect on the body. Allium Mongolicum Regel polysaccharides improved T-SOD, CAT, T-AOC, GSH-PX in serum, liver, kidney and heart and it could reduce the content of MDA. Compared with the blank control group,①In serum and liver, T-SOD of 800 mg/kg ? BW of the Allium Mongolicum Regel polysaccharides was the strongest (P﹤0.05); In kidney and heart, T-SOD activity of 400 mg/kg ? BW of the Allium Mongolicum Regel polysaccharides was the highest (P﹤0.05).②In serum and tissues 400mg/kg ? BW of the Allium Mongolicum Regel polysaccharides can be best to decrease MDA. In addition to the heart of the difference was not significant(P﹥0.05), the other differences were significant (P﹤0.05).③In serum and liver, 400mg/kg ? BW of the Allium Mongolicum Regel polysaccharide was the best to increase CAT(P﹤0.05); In kidney and heart, 800mg/kg ? BW of the Allium Mongolicum Regel polysaccharide was strongest. In kidney,CAT activity was significantly different (P﹤0.05), but the difference was not significant in heart (P﹥0.05).④In serum and heart, 800mg/kg ? BW of the Allium Mongolicum Regel polysaccharides had the best effect to increase the T-AOC(P﹤0.05); In liver, 200 mg/kg ? BW of the Allium Mongolicum Regel polysaccharides had the best role(P﹤0.05); In kidney, 400mg/kg ? BW of the Allium Mongolicum Regel polysaccharides was the best(P﹤0.05).⑤In serum, 800mg/kg ? BW of the Allium Mongolicum Regel polysaccharide efficiently enhanced GSH-PX activity (P﹤0.05); In liver, GSH-PX activity of 200mg/kg ? BW of the Allium Mongolicum Regel polysaccharide was highest (P﹤0.05); In kidney and heart, 400mg/kg ? BW of the Allium Mongolicum Regel polysaccharide improve GSH-PX activity efficiently(P﹤0.05). In conclusion, 400mg/kg ? BW of the Allium Mongolicum Regel polysaccharide goup had better antioxidation than other groups in vivo.
