Using AFLP markers, selected out four pairs of the selective amplification primers of high polymorphism, well-distribution and strong resolution, analysis on genetic diversity of three Staphylea bumalda DC groups (Shitai in Anhui province, Dawu in Hubei province, Tongbai in Henan province) and three Elaeagnus mollis Diels groups(Sanmen,Xiangning and Yicheng in Shanxi province).1) Amplified a total of 489 bands of Staphylea bumalda DC, including 460 bands of polymorphic, the percentage of polymorphism was 93.99%. Number of alleles (Na) was 1.9407, Nei's genetic diversity index (H) was 0.2190, Shannon's information index (I) was 0.3493; Amplified a total of 372 bands of Elaeagnus mollis Diels, including 348 bands of polymorphic, the percentage of polymorphism was 93.55%. Number of alleles (Na) was 1.9355, Nei's genetic diversity index (H) was 0.2204, Shannon's information index (I) was 0.3498. Those showed genetic diversity of Staphylea bumalda and Elaeagnus mollis was abundant.2) The results of Shannon's information index,Nei's genetic diversity index and genetic coefficient of variation (Gst) showed: Population's genetic variation of Staphylea bumalda and Elaeagnus mollis existed within every group, groups of genetic variation were not significant differences.3) Nei's genetic identity of Staphylea bumalda groups was between 0.9649~0.9763, the genetic distance was between 0.0240~0.0357. Nei's genetic identity were the highest between the groups in Dawu and Tongbai, Nei's genetic identity were the lowest between the groups in Shitai and Tongbai, Nei's genetic identity of groups in Shitai and Dawu were in between. Nei's genetic identity of Elaeagnus mollis groups was between 0.9821~0.9893, the genetic distance was between 0.0108~0.0180. Nei's genetic identity were the highest between the groups in Yicheng and Xiangning, Nei's genetic identity were the lowest between the groups in Yichengand Sanmen, Nei's genetic identity of groups in Xiangning and Sanmen were in between. The results of UPGMA cluster analysis using genetic distance to the groups of Staphylea bumalda and Elaeagnus mollis, which showed that its had close relationship between the various groups.
