Two heading Chinese cabbage(Brassica campestris syn. rapa L. ssp.pekinensis (Lour) Olsson)'08S555'and'shanqiubai'provided by Chinese cabbage research team of Northwest Agriculture and Forestry University were the experiment materials in this study and the factors affecting in vitro regeneration of cotyledons of them were studied first. The Gene of abnormal floral organs transformation factor was constructed as sense expression vectors and transferred to'06J28'with the method of our pro-researches, and the delay screen procedure and effect was studied especially. The results are showed:1. The results show that either the combination 6-BA and NAA or TDZ and NAA,'shanqiubai'always had higher regeneration ratio and lower regeneration coefficients than'08S555'2. In the 6-BA and NAA combinations condition:'08S555'got the highest regeneration ratio of 28.33% and regeneration coefficients of 2.81 at 5~7 mg?L-1 6-BA and 0.3 mg?L-1 NAA level ;'shanqiubai'got the highest regeneration ratio of 68.33% and regeneration coefficients of 1.09 at 5~9 mg?L-1 6-BA and 0.5 mg?L-1 NAA level. In TDZ and NAA combinations condition:'08S555'got the highest regeneration ratio of 45.83% and regeneration coefficients of 2.55 at 0.5~1 mg?L-1 TDZ and 0.5 mg?L-1 NAA level;'shanqiubai'got the highest regeneration ratio of 79.17% and regeneration coefficients of 1.24 at 0.1~0.7 mg?L-1 TDZ and 0.5 mg?L-1 NAA.3. When 5 mg?L-1 6-BA,0.5 mg?L-1 NAA and 6 mg?L-1AgNO3 added in medium, the highest regeneration ratio of'shanqiubai'is 74.25%; while TDZ (0.3 mg?L-1), NAA (0.5 mg?L-1) and AgNO3 (6 mg?L-1) added in medium. regeneration ratio of'shanqiubai'reach 87.50%.4. NAA added in 1/2MS medium had better effect on root growing than IBA. At the concentration 0.3 mg?L-1 of NAA, the seedings had developed good root system with more and thick main roots, as well as thick fibrous roots.5. Five transgenic plants of'06J28'with target gene was obtained by PCR test.
