Bromeliads, the excellent indoor ornamental foliage and flowering plants, its consumption rose sharply in recent years, while there are limited resources in China. The introduction of foreign genes by genetic transformation provides an alternative approach to enrich germplasm resources.In this study, we evaluated a germplasm collection and established the particle bombardmented-mediated transformation system for Bromeliads.More than 661 germplasms were obtained and evaluated, among these Guzmania 'Diana'and G.'Focux'were donated as transgenic materials.The high efficiency DNA extraction method was established through extraction liquid, material, grinding methods, settling time, and so on in different bromeliads species.We also established the efficiency micropropagation system throught tip and sheath of Guzmania'Tutte Frutte'. The results indicated that the best induction medium was MS+6-BA0.5mg/L+NAA0.5mg/L, the best differiation medium was MS+ 6-BA2.0mg/L+NAA0.5mg/L+2,4-D2.0 mg/L and the best root induction medium was MS liquid medium.The study of microprojectile bombardment-mediated genetic transformation for Guzmania was carried out with the shorten stem of'Diana'and'Focux', and three positive lines were obtained and confirmed by PCR. The expression of pBIN438-GFP in the two kinds of Guzmania leaves was observed under the Laser scanning confocal microscope. The results implied that the microprojectile bombardment-mediated genetic transformation system for the ornamental bromeliads was established, and provides an alternative approach to enrich ornamental bromeliads germplasm resources.
