To study the pharmacokinetic of Chinese traditional medicine "LH" main component in rats, at the fiirst, we should extract main component berberine of rhizoma coptidis in this test, abstract and purify it with column chromatography fractionation, use HPLC to assay the purified product.. Using rope extraction process to extract main component forsythin of forsythia suspensa, screening optimization react parameter of rope extraction process according to the L9(34) orthogonal test table. Abstract and purify the forsythia suspensa extraction of the best extractive technology of rope extraction process with versamid adsorptive process, use HPLC to assay the purified product..Use HPLC still to assay caffeotannic acid of flos lonicerae to keep this test regularity. Simultaneous determination three main component (berberine, forsythin, caffeotannic acid)of Chinese traditional medicine "LH" with HPLC.To research the pharmacokinetic of Chinese traditional medicine "LH" (caffeotannic acid, forsythin, berberine) in rats.3p97 procedure was used to handle time-dependent state of aforesaid three component in rats blood plasma, and used compartment model (one-compartment, two-compartment,three-compartment) to simulate differently, we could procure fig. of AIC and coefficient correlation, obtain pharmacokinetics parameter according to the principle of AIC fig. minimum and r fig. maximum. Analyses distribution condition of three component (berberine, forsythin, caffeotannic acid)of Chinese traditional medicine "LH" in rats several organs, dejecta and urine with HPLC.We can decide the most superior response parameter of the rope extraction process is A2B1C3D3 according to statistics analysis. The purity of berberine in rhizoma coptidis, forsythin in forsythia suspensa and caffeotannic acid in flos lonicerae reach differently 95.21%,95.012%,87.16%. The purity of three component (caffeotannic acid, forsythin, berberine) in "LH" of tested solut reach differently 14.468%,27.709%,15.312% with HPLC in this methord. The pharmacokinetic proceeding of three component in rats are linear in the dosage range of reserch. Distribution state of three component are general in rat liver, lung and several digestive, while minor in other organs. In liver, the concentration of caffeotannic acid is the topmost. And the content of three component is little in dejecta, is the lest in urine.The purity of forsythin and berberine reach 95.012%,95.21%. The concentration of berberine, forsythin and caffeotannic acid are 15.312%,27.709%,14.468% in rats blood plasma with HPLC. Berberine and forsythin were fitted to be a one-compartment model, caffeotannic acid was fitted to be a two-compartment model.
