Studies On The Domestication Cultivation, Anti-Hyperglycemic And Antioxidative Effect Of Inonotus Hispidus

The medicinal fungus Inonotus hispidus extracyed from mulberry in southern Xinjiang is used as the basic experimental materia and the optimum artificial domestication cultivation conditions have been determined and the chemical composition,antidiabetic and antioxidative activities have been preliminarily studiied.The result indicated that the optimum temperature for Inonotus hispidus is between 25℃-35℃,the pH value 7-9 and C/N 90:1-80:1. When the lime composition is1.5%,the pH value can be altered relatively and the trace elements can be provided to facilitate the growth of the mycelia.The optimum composition of the ingredients for the Inonotus hispidus in southern Xinjiang is: Cotton Seed hulls 92﹪,bran 5﹪,gypsum 1﹪,oil residue 1﹪, lime 1﹪.The wild Inonotus hispidus group,artificially cultivated group and liquid fermentated mycelia group have been preliminarily studied by comparison and the result showed that the above-mentioned three groups all contain alkaloid,saccharide and polysaccharide,sterol and triterpenoid,saponin and glycosides,proteins,amino acids, etc.Through single-factor experiment and orthogonal experiment, the optimum process conditions for the extraction of Inonotus hispidus is:temprature 80℃,extraction time 2h,ratio of solvent to solute 1:40, lixiviation time 3, the composition of polysacchride 86.2mg/g.The above-mentioned three groups were processed with 75% cold ethanol and lixiviated for three times and 7d per time. After decompression and condension the product were extracted successively with petroleum ether,chloroform and ethylacetate and the products in proper oder are petroleum ether extraction,chloroform extraction and ethylacetate extraction.The trial drug for the three groups of Inonotus hispidus is polysaccharide,the blood sugar level of normal mice and tetraoxypyrimidine-infected mice indicated the trial drug cannot affect the blood sugar level of the mice sinificantly. 21 days after injection of the drug, the tetraoxypyrimidine-infected mice`s blood level have been reduced to a certain extent and moreover, the wild high dose group(200 mg/kg weight) and medium dose group (150 mg/kg weight) showed a significantly better hypoglycemic effect than the high dose artificially cultivated group and liquid fermentated mycelia group.The removal ability of the extracts to DPPH free radicals,hydroxyl radicals and oxygen free radicals has been measured by DPPH method, salicylic acid method and pyrogallicacid method. The result showed that when the concentration was 10mg/ ml,the removal of DPPH free radicals is good and the polysaccharide composition of the wild group can reach to 95.82%,the cultivation group 95.82%,liquid group 83.48%,enjoying a more effective removal efficiency than other groups. The removal efficiency of hydroxyl radicals is 92.60% of wild group, 91.21% of cultivation group and 85.29% of liquid group, enjoying a more effective removal efficiency than other groups. The removal efficiency of O2- free radicals is 92.94% of wild group, 89.15%of cultivation group and 82.36% of liquid group, enjoying a more effective removal efficiency than other groups. The removal ability of the three groups is wild group>cultivation group>liquid group.The study of the antioxidative activity of Inonotus hispidus in the mice shows the wild group and cultivation group performs better than negative control group and worse than the positive control group in the removal ability of free radicals from liver and blood of the mice, in improving SOD composition and in reducing MDA composition, possessing great antioxidative activity.
