| Anguilla Anguilla is an main aquaculture species in our country.With the development of the artificial cultivation, there has been a trend of a spread of the diseases of Anguilla Anguilla.The morbidity and the mortality of the diseases have gradually rised,which has been one of important constraint factors to the development of the culture of Anguilla Anguilla in our country.There are many species of pathogens which can harm Anguilla Anguilla though the prevention and cure technology of the diseases has lagged, especially the technology about virus diseases. People have a simple comprehension of the virus diseases of Anguilla Anguilla.The main reason has constrained the thorough research of the virus diseases of Anguilla Anguilla is the lack of the adaptive cell model. So far, there has been not any report about Anguilla Anguilla cells at home.To establish a cell model used to isolate and identify the virus diseases ,and to make a thorough investigation about the prevalence and the transmission of the virus diseases, the pectoral fin cells of Anguilla Anguilla (the 21st generation) were studied in this study. The cells were cultured by artificial mediums in vitro.This study researched the best conditions of the pectoral fin cells of Anguilla Anguilla cultured in vitro.On the basis of the best medium, basic fibroblast growth factor (bFGF) and insulin-like growth factor-1 were added to observe the effects of bFGF and IGF-1 on the proliferation of the pectoral fin cells of Anguilla Anguilla.The pectoral fin cells of Anguilla Anguilla were observed and compared ,while the cells were cultured under different mediums (L-15,RPMI-1640,DMEM),different serums(BS,FBS),different serum concentrations(5%,10%,15%,20%).In certain time,the cells were collected and counted ,then the growth curves of the cells were drawed. The effects of the different conditions on the proliferation of the pectoral fin cells were evaluated by the growth curves.The results showed tha L-15 was the most suitable medium ,while RPMI-1640 took the second place and DMEM was the poorest;FBS was better than BS,while 10%FBS was the best concentration.The effects of bFGF and IFG-1 on the proliferation of the pectoral fin cells were observed so as to evaluate whether bFGF and IFG-1 had promoted the proliferation of the cells.The results showed that bFGF had an obvious effect on promoting the pectoral fin cells with L-15(10%FBS),and the best concentration was 10ug/L;however IGF-1(from 5ug/L to 50ug/L) was not conductive to the proliferation of the pectoral fin cells.The study found out the best conditions of pectoral fin cells within three mediums,tow serums and tow growth factors.The study preliminarily confirmed that bFGF had an effect on promoting the pectoral fin cells while IGF-1 was not conductive to the proliferation of the cells.The results provided a practical and effective formula and culture method that can reduced the doubling time and increase the cell number in certain culture period.The method showed potential value in pathogens isolation and identification of the virus diseases and vaccine preparation. |
