In 2009, a kind of bacterial disease in processing tomato was observed in Inner Mongolia Bayanaoer, 7 strains were isolated from diseased specimens, 7 strains were inoculated on tomato and same symptom appeared as naturally infected plants, the bacteria were isolated again from inoculated tomato. By morphological characterization, culturing patter, stain reaction, physiological and biochemical reactions, 16SrDNA sequence analysi, pathogen were identified as Pseudomonas syringae pv. tomato .The antiserum of cells of PST and the antiserum of whole protein of PST was prepared in this study.The result of antiserum titer by Tube agglutination has been reached 1:1280.The speciality of antiserum detected by Immuno-isolation and Tube agglutination showed that the speciality of two antiserum are consistent.PST is positive, Pseudomonas syringae pv.lachrymans and Pseudomonas fluorescens are both negative.The result showed that the speciality of two antiserum are higher. Immuno-isolation simulated the detection of bacteria in seeds,the precision of bacteria detection to app. 0.8×102 cfu/ml.PCR simulated the detection of bacteria in seeds, the precision of bacteria detection to app. 2.8×105cfu/ml.
