Spit liquid is produced by Bombyx mori during their eclosion stage.It softens and dissolves cocoon layer so that the adult can escape from the cocoon. In order to obtain the function of secretion organs of Bombyx mori' s spit fluid,we will combination physiology experimental method and proteomics research in this theses.We will research the activity of spit liquid through removed the first maxilla and salivary glands. We will research the protein of spit liquid Through MALDI-TOF-MS and LC-MS/MS methods. We will discuss the function of the first maxilla and salivary glands during the synthetic process of spit liquid.Our research lay the root for the synthetic mechanism research of cocoonase and the protein research of spit liquid.1. First maxilla and salivary glands is not very obvious effect to the development process between pupa to moths, the pupas almost can development moths after remove the first maxilla or salivary glands, the reason of death is that the pupa will lost much blood after remove the first maxilla or salivary glands, also the pupa may be infection easily after the pupa have wound.2. We have assaied the sericinase, protease, esterase and fibrinogenase activity of the first maxilla crystal, crop liquid and the spit liquid after remove the first maxilla or salivary glands.it is indicate that the cocoonase in spit liquid is only come from the first maxilla, the midgut also can secrete the cocoonase,but the cocoonase in midgut will not participate in the process of taking off from cocoon, the activity of the cocoonase in midgut will lost when the spit liquid get to the in vitro. The extensional mechanism why the activity of cocoonase in midgut will lost is not very clear, but the experimental result make clear that it's happen due to the salivary glands.the solution in midgut can provid a suitable pH.salivary glands is very important in the process that the moths take off from the cocoon, it can regulate the activity of cocoonase.the midgut can secrete another enzyme that have fibrinogenase activity.3. We have identifid the SDS-PAGE different points.The comparison to the moths that remove the first maxilla have a different point, it is cocoonase, there is not a protein strap in the same position in the SDS-PAGE of the spit liquid after remove the first maxilla, so we conclude that there is not cocoonase remove the first maxilla. In other words, the activity of the cocoonase in midgut have lost the spit liquid get to the in vitro.4. We have obtained 29 proteins in the spit liquid thongh LC-MS/MS assay,the enzyme possess about 63%,it also include transferrin, immune-related protein and serpin.the serpin may be from salivary glands.
